      I have been meaning to make a video of my whole sub prep. Thank you for reminding me. I basically use 80% coir, 20% verm by volume. Then I load it into a big pot, add an excess of water, cover with lid, and boil/steam for 30 minutes. You don't have to soak or add exactly the right amount of water. Just enough to hydrate it. I then squeeze it a handful at a time when I mix with spawn.

    Dr. Ed, I graduated from university almost 20 years ago and rarely have the opportunity to ask advanced science questions to knowledgeable people such as yourself.
    I never realized how valuable an experience and opportunity it is to be able to get high level answers from PhD-educated scholars. The older I get, the more I realize how little I understand. So thank you for helping demystify the world little by little.
    Even though it won't get you the most views, I hope you're still open to doing some videos about a bit more advanced topics on genetics, breeding, etc. (e.g., how does Monsanto track whether their seeds are being used by farmers to "illegally" breed successive generations; how does Monsanto prevent you from being able to grow plants you get from the store; how does environment affect gene expression; etc.)

    Do you sit on that couch behind your flow hood? Also, why are your cultures in a tent? does that provide anything extra besides just shelving?

      The couch came with the house. I don't use it. Tent is just a slightly dirty/old Martha. Just use it for shelving now.

    @Edward Grand Could you do a video like an instruction video that I could follow using your Great White Monster and Bee Pee swabs showing your recommended technique using your swabs PLEASE. I get confused like am I supposed to grab drag , do a S swipe or a Z swipes or maybe a geto swipe 🤷‍♂️ There are so many that seen you do and I'm excited to use your genetics and I don't want to mess it up.

      I use the 'grab and drag' technique with all my swabs now. It is superior to all the other methods I have seen. Make sure you get the spores/cotton piece jammed into the agar crevice. That is the key to it's success.

    Is there ever an instance when two monos will not create a clamp?

      Yes. If they have the same mating alleles (or they are contams). I have found almost 100% compatibility with monos that are not from exactly the same cultigen/parent. I don't worry about it anymore. If you have two clean monos, they will almost always make clamps. If they don't, something else is wrong.

      Yes. I plan on doing some videos that are literally just me working, setting up, etc. in real-time with the camera running. I might even try to answer some questions while I do it. I think people really need to get an idea of the time frame and actual work instead of a 10 minute edited video that took place over several days or weeks.

    Can you revive a culture from a dried mushroom? Maybe from spore though it may require a lot of plate work?

      Yes. It sometimes works. Depends on the original drying temperature and the way it was handled. Use bleach, peroxide, and water to prepare the sample. There are a few ways to do it.
      You could also just try getting spores from gill tissue. This has similar issues (age and temp), but might be a better approach.

    Just had my TAT x Enigma blob spores from you germinate! Really excited to see what f2 and f3 generations hold for me! I have a question that might be good, can you do a short video or segment on germinating stubborn spores and possibly methods to get it done. ive been struggling a lot lately with some galindoi spores that ive had for almost 3 years and cant seem to get germinated. I tried re-hydrating them using the pressurized syringe method, will be interesting to see if it works.

    Your prep work that leads up to sitting in front of the FFU in aseptic environs. Takes me about 20minutes and not sure if I've missed any keys steps. Don't have to see underwear... but arm washing, gloves, mask?, wipedown-where and with what, how to get tools clean ready and in place, petri treatment so that labels aren't washed off but they are clean, on and on. Do you spray front of FFU? Wipe from FFU outwards? Keeping it clean while you work - what can you touch or should not touch (dirty) or when to spray gloves again - aseptic technique and things to keep in mind as you work so that you don't contaminate. Do you clean your phone in case it rings or alarms while you're working? If you reach outside the flow but only set things down, not touching any unclean surface, can you continue to work or should you spray... blah blah blah

      Thanks for the suggestion. Here is my recipe.
      GRYA recipe
      3% Glutinous Rice/Whole wheat flour
      2% Agar
      0.1% Baker's or Nutritional Yeast
      Food Coloring (if desired)
      FOR 500 ml Total volume
      15 g Glutinous Rice/Whole wheat flour
      10 g Agar
      0.5 g Baker's Yeast
      Food Coloring or 1g charcoal if desired.
      500 ml tap water
      - Should make between 20-25 standard size plates.

    Does LC work as long term storage the same way as you demonstrate with the water technique? And why would it not? I think I have some ideas with that. What is your take?

      Yes. I have pulled viable culture from 2 year-old LC that hasn't been agitated or moved. I use LC as a sort of back-up of a back-up for cultures. Or you can just make syringes and store them in the fridge. These might last for years.

    Recently rather than mould, I seem to have consistent issues with bacteria (using flowhood, sterilising in an autoclave for 3hrs)... I get some jars looking partially 'ice creamy', often with a bit of an odd smell - still produce fruits but just annoying that it keeps happening.
    Any suggestions?

      Check the cultures on agar. Do they have a halo near the growing edge?
      Keep one jar of grain uninoculated to see if it goes bad.
      Try not to overwater/mist.
      Squeeze your substrate to Field Capacity.

      #7 scalpel handle and #11 blades. That is the only thing I will ever use again. The handle is slim and long keeping your hands away from the work. The #11 blades are thin and sharp, making it easy to work with. I have tried a lot of alternatives. None of them work as well as this combination for me.

      Paddy rice. It is the actual rice they use for planting with the husk/hull still attached. Cheap AF here. They use old 'human' rice for dog food because it's basically cheap and tastes not so good after a year of storage.

    Congratulations on the new flowhood! When I started using my first flowhood - I loved the freedom of movement (and of course the much much lower contamination rate. Important to note that good sterile technique is still important in front of a flowhood!
    Here is a question for you (and probably relevant given the location of your hood), what is the best way to protect the HEPA filter from airborne contaminants while not in use? Also - how frequently (as a function of bourse of flowhood use) do you change your intake filter?

      @@edwardgrand pre filter is a very good idea for extending the life of your main filter … but you already know that, I suspect. My pre filter gets a pretty good layer of cat hair and lint on it and seeing it there makes me feel better about where I can’t see 🤪

    man, i have a question, i just thought that i could keep my culture on grain and have a master jar and only do g2g, so that would mean no lc bullshit anymore so i was wondering why people dont do that, how long could i keep doing it with a single culture in a single jar, if i put it in a new jar would it keep on living infinitly blah blah blah, anyway just asking for a video about cloning/keeping a culture via G2G transfer, thaks man your the best frl

      I don't like doing G2G. It is kind of dirty, and the spawn will eventually stop performing well. I stick close to my spores and original cultures. I usually only run a new cultigen once or twice, so it's pointless for me to do G2G. In theory, you are 100% right. I'm not sure why people don't do it?

      I will never be getting into P. semi. They are a low-yielding outdoors species that would be very difficult for me to grow indoors here in Thailand. I have my hands full with the cubes. Probably enough work for about 10 lifetimes sitting in my front room. Almost overwhelming at times. I'll leave the semi for someone else.

      They both work fine. I have used both and many models and it is all personal preference. I like the horizontal (like the one in video). The name refers to the flow of air. Confused me for a while.

      @@Deltaforce650I have seen people do that. The only issue would be the fan and if it can handle the different orientation. The FFUs are meant to be in the ceiling, but the fans don't seem to mind being vertical.
