Thanks for this vid, best Pandemic Iberia Video out there! There's not a good video for step by step setup and run through out there thats as clear as yours. Others always refer you to the original Pandemic which is bad for players who just got into this series.
and what about the purification action, can we discard any colour in the particular region to add purification to that region. or it should be the colour on where we currently at ?
It has to match the color of the city you're in. However, when you discover a cure, you can use those color cards to purify regions as well as the color of the city you're in
Everything else is cleared, I had seen rahdo’s video and other board game reviewer’s video They in their video said we can purify by using the colour of the cities int hat region. Say we have a region which has 3 blue cities and 1 red and we are currently on a blue city , we can discard a red card to purify that region . Rahdo iberia gameplay runthrough 10:45
also if Lisboa has 3 disease cubes and we decide to discard Lisboa card and building a hospital, will that action cause to remove the disease cubes as well?

It will not. You will still have to take actions to remove the cubes
