For anyone trying to get all 16 combat variations (note that this is separate from the gadget variations): 1. Strike 2. Redirect 3. Counter 4. Stun 5. Super Stun 6. Aerial Attack 7. Beat Down Finisher 8. Critical Beat Down Finisher 9. Ground Pound 10. Aerial Juggle 11. Blade Dodge Takedown 12. Special Combo Takedown 13. Special Combo Disarm & Destroy 14. Special Combo Multi Ground Takedown 15. Environment Takedown 16. Evade Charger (Thugs would have the red warning and charge at you e.g., see 0:16. BUT, you can't use the Batarang to knock them down. Instead evade the charger to get this variation)
I've tested it and evading a charging thug does not seem to count as a variation it counts as redirect but throw counters and regular counters are counted separately
It's an upgrade called Disarm and Destroy, available in the combat tree, you unlock it during the campaign and it costs 6 Wayne Tech Upgrade points if I remember correctly, the combo can be performed by pressing Square + Triangle at the same time
Altogether with one finger, it might be tricky to find a comfortable way to press them at the beginning but you'll get the hang of it eventually, keep in mind that only Batman, Azrael and Batgirl can perform Disarm and Destroy
In challenge mode I think this video has all of them for combat, 16 for combat + 10 for gadgets = 26 total In campaign the number of variation bonuses is measured differently so it can go up to 20+ for combat

do you get more points on the end screen if you play as a duo and use both the second characters unique movies and the dual team takedown?

It does, you have the Tag Team Takedown + 2nd character variations, achievable in Guardians and Gotham Knights combat challenge
