      For recreational use, it's already easy to legally get spores online and get to growing. This kind of stuff is more for mass production to enable low cost use in therapeutic settings.

      @@orion9k Well to get psilocybin you don't even have to grow anything yourself. Just go to your local park and look for naturally growing Psilocybe-species, like P. semilanceata. They are pretty widespread and contain high concentrations of those active compounds. Just remember to not eat any mushrooms that you're not 100 % sure what it is.
      So yeah, getting psilocybin is not a hard task and anyone could do it.
      But there is a big difference between growing your own mushrooms from mycelium and grains and putting some yeast in a bucket with water and sugar and getting psilocybin out of it. And last time I checked there wasn't any vendors offering these kinds of yeasts for sale, so brewing your own psilocybin is still a thing of the future.

      Most bakers use a single strain of yeast for each type of bread. The same scrutiny is exercised for beer and wine as well as mashes for spirits. There are other situations where it would slip on there. Sourdough bread is usually a local yeast bouquet from putting a dish of water with a touch of flour out, then when the tase is preferable, the "mother" batch is kept and regrown in a dish with a cloth over it to keep other spores out. Occasionally, they will split the mother and allow more local yeast to settle in. In that case, you could definitely affect a sandwich. Some fruits are fermented with local yeast as well to produce wines. If you have ever seen the video of the chateau elan reporter (ooh oooh ooh ooooh) making wine, you see her smash grapes with her feet. Think about that, your feet generally stay warm, dark, and often moist, making a perfect habitat for yeast to live. There are other places on the body susceptible to yeast reproduction. Think infections specifically in the downstairs regions. Now imagine a foot colony or other colony on your body producing a constant supply.
      Now imagine how long it would take the general public to figure out they aren't just tripping over their feet, but tripping because of them.

      nah unless you intentionally try to do it, giving yourself auto-brewery syndrome is pretty hard. The body becomes tolerant to this drug insanely fast too so I can't imagine the effects would really last all that long even if you did succeed

    Very very nice video, it's not easy to learn BioTech (or not as easy as many other topics) by yourself and you made a amazing job explaining this paper. Had a little ChatGPT window to get some definitions or insights on things you mention I didn't know or remembered.
    Also 100g equivalent of dry mushrooms in a 1L bioreactor is insane !

    BTW - a bright red dragon told me getting the dose right is a bit tricky. A lot of plants produce DMT so maybe those pathways are known and that molecule will be next. As a Chemical Engr. I see DMT as better suited to extraction, purification, analysis, quantification and storage for the home craft brewers. Now you've got me thinking about how to process the Psi-brew. Thanks for this video, and congrats on your PhD.

      @@microfx Structurally yes. Purification, isolation & preservation require higher skills. DMT processing is a known art. Having said that it will be interesting if the yeast method gets out in gen pop and producers find a product form that meets consumer requirements. I'm betting the community works out those challenges.

    There's a paper from around 15-20 years ago on making hydromorphone from yeast that I've been waiting to see actual usage. A video on that though similar would be interesting too. The paper was widely reported at the time but I haven't heard anything since.

      opioid production in microbes has had a big push recently, but as far as I know, titers are still in the ug/L range, which may make the process not very economically viable yet. I did find some older papers on opioids and microbes, one where they fed a bacteria morphine as the sole carbon source?     Brewers yeast will reduce tryptaphan to tryptaphol as it is, then put an enzyme coded to attach a dimethylamine tail on it. At that point you got dimethyl tryptamine. For psilocybin you would want the enzymes from the fungi and increase the free phasphate added as diammonium phosphate to push the equilibrium toward the production of psilocybin.

    Can you please make a DIY tutorial for making galactosyl-lactose (aka beta-GOS) at home? Evidently certain kinds of beta-galactosidase enzymes under certain conditions can cause partial hydrolysis of lactose and also combining of galactose with lactose to form galactosyl-lactose. Beta-GOS is the best prebiotic in the market and the literature for improving lactose tolerance.

      Is casomorphone addiction the reason people that are lactose intolerant won't give up on dairy? Similarly, if I had celiac, I wouldn't eat wheat, not look for more ways to manipulate it so I could eat something harmful, as a drug. Guess that's just me being too logical again, sigh.
      If a "food" made me sh*t my brains out, I probably would take the hint and avoid it, but that's me.

    i got a general question on such engineered yeast strains: do they diverge from the engineered variant after a while? is there a contamination risk with wild yeast strains etc.? i have no idea about any genetic engineering/biotech topics, i just find it fascinating and also saw some simpler DIY stuff about it on the thought emporium which was interesting to see how it works in practice. i'm wondering because i've heard about these types of yeasts quite often but i'm wondering if it could ever happen that someone reproduces one of these papers and just starts selling this stuff 🤣or if that's unreasonable because it'll degrade too quickly

      People could definitely make this and sell it! Yes, over time the genetics of the strain will change. It is not evolutionarily advantageous to have these bulky, irrelevant genes integrated, so the yeast will eventually lose them given enough generations. However, we can create glycerol stocks (there's a video on this on my channel!) of the yeast to basically freeze them in time. It's like finding an heirloom tomato seed from a 200 year old tomato or something. The genetics will be very similar to the first sell that had those genetics.

    That's nice and all but how do you purify it? You're going to have to separate out the product from yeast culture.
    Do you purify with a large column? What kind of column do you use to purify it too? I would be interested in seeing a paper purify a small molecule product like that from cell culture.
    Can you use an instrumental separation technique to purify it from cell culture in fraction pooling?
    Like these are the things I'm curious about lol.

      Yep! Brewer's yeast is the most well studied yeast and thus the easiest to work with (also very high homologous recombination efficiency).       @@DIYBiotech Yeah that high HR efficiency must make it an easy choice for this kind of work. Always nice when something like that appears in nature. Kinda like how P. Cubensis has the good fortune of being so easy to cultivate on so many different substrates. Little serendipities, I guess!
      Gotta say though, in dissent with your opinion on the inclusion of Methods and Materials sections, I rather like seeing them, not just for more intensive peer review and replication, but for transparency and the democratization of science as well. Certainly a specialist might be able to infer the process from the rest of the paper or even contact the authors, but I like the idea of non-specialists being able to either replicate the results or concoct offshoot experiments based on the specific methodologies employed. I mean, we wouldn’t be able to have the “will someone try this at home?” conversation without it. Moving away from them feels like a “pay no attention to the man behind the curtain” moment, and even with the best of intentions still might come across that way. I’d think, especially given the current academic climate, we’d want to reduce that kind of academic stratification at any opportunity, even if the inclusion of that information may be redundant for certain parties. I’m interested in what your rationale is with regard to not liking them. Maybe I’m missing something?

      The strain from the paper probably makes some alcohol, but likely less than 1%. You could start with a strain that makes a lot of alcohol and modify from there instead! Not all S. cerevisiae make a bunch of alcohol.

    Why would you not want materials & methods in the study? How would peer review work? How would you check that their experiment wasn't poopoo?

      Great question! When we peer review a paper, we don't actually recreate any experiments, typically. We just take their word for it. I wasn't super clear here, but the only time we use materials and methods is if there is a small part of the experiment we want to copy for our own papers we are working on. It is more common that people want to just read the results. We usually don't have time or don't care about the minute details on how they got their data!

      Could definitely do it chemically! But maybe there's an enzyme out there that could make this... Or maybe use a different precursor and hope the enzymes in the pathway are promiscuous to it.

    Sorta related to this, it's probably just fiction but I just started watching the new Penguin TV show & they supposedly developed a "new drug" and it surprised me when they showed a chemist, mycologist who found a way to grow bleeding toothed fungi that ooze a red fluid drops 🩸 🍄 and they extract a psychoactive drug from that red fluid that's different than the usual 2 compounds you usually get from psychoactive fungi. I know it's probably just made up movie BS but it still interested me because I know those bleeding tooth fungi are real. I wonder if anything can be utilized thru that?

      This could realistically be done in the next decade. Not the most practical way to make chemicals, but it could be done with relative ease.
