ഒരു Bacterial cell പുറത്ത് നിന്ന് ഒരു DNA യെ (eg: Plasmid) ഉള്ളിലേക്ക് എടുത്താൽ അവയെ Transformants എന്നു പറയാം. ഒരു bacterial cell ഒരു rDNA യെ (eg: Plansmid with gene of interest) ഉള്ളിലേക്ക് എടുത്താൽ അവയെ recombinant എന്ന് പറയാം
ബാക്ടീരിയകളെ Petric plate ലാണ് വളർത്തുന്നത്. Product ലഭിക്കാൻ വേണ്ടി നമ്മൾ ബാക്ടീരിയകളെ വളർത്തുന്ന petric plate നെ Master Plate എന്നു പറയും. ഇതിൻ്റെ ഒരു കോപ്പിയെ Replia plate എന്ന് പറയും.
Replica plate ൽ tetracycline എന്ന antibiotic ഒഴിക്കും. അപ്പോൾ recombinants നശിച്ചുപോകും. കാരണം അവയിലെ tetR gene ൽ foreign gene ഒട്ടിച്ചതിനാൽ tetracyclin എന്ന antibiotic നെ പ്രതിരോധിക്കാനുള്ള കഴിവ് നഷ്ടപ്പെട്ടിരിക്കുന്നു.

Cloning vector പ്രത്യേകം ഉണ്ടാക്കുന്നതാണ്. അത് host bacteria യിൽ നിന്നെടുക്കാം അല്ലെങ്കിൽ അനുയോജ്യമായ മറ്റു ബാക്ടീരിയകളിൽ നിന്നെടുക്കാം
