My teacher told us (1 minutes MB, 2-5 minutes of iodine, 6 seconds of ethanol and then 2 minutes of saffranin) and I spent the whole lab yet can only achieve all purple :( so Im now wondering if i put the stains on too long and then the ethanol not enough. 🤷🏻♀️
jennifer pan it usually means you didn’t run the enough ethanol so next time run it till you don’t see any blue color at the bottom and immediately rinse

Yes we have to wait orelse the inoculation loop which was incinerated,it will be hot so when you keep that loop in the bacterial culture there may be chance in which bacteria may die.

You do not need to let it air dry. You do the heat fix to not only dry it but it kills the bacteria and makes it stick to the slide.

Not really 10 seconds, as long as you test it on the agar, it doesnt burn the agar it should be cool enough to extract a bacterial culture
