Tryptophan Metabolism (Degradation) and the Kynurenine Pathway

  • Lesson on Tryptophan Degradation (Metabolism) and the Kynurenine Pathway. Tryptophan is an essential amino acid required for the production of serotonin and melatonin. Tryptophan can be degraded by the kynurenine pathway, which can be activated by stress and immunocytokines. Activation of the Kynurenine pathway can reduce the production of serotonin. Kynurenine metabolism will eventually lead to the production of the end metabolites quinolinic acid and picolinic acid. Quinolinic acid and the Kynurenine Pathway have been implicated in various psychiatric conditions including major depressive disorder, due to the reduction in serotonin production and function.

    Super cool to know that tryptophan + inflammatory cytokines or + stress (cortisol) = kynurenate acid / quinolinic acid (which is neurotoxic/can cause depression), and lead away from serotonin (feelings of wellbeing) and melatonin (better sleep). Biochem incentive to reduce inflammation and stress :)

    This is the very fist channel that actually explains metabolic pathways in an understadable and complete way. Several methabolic pathways I have had researched yet didnt understand/find here they are. Its lovely (greetings from mx)

    Thank you so much sir for this amazing video. Could I ask about NAD (niacin) please, what I got from your explanation is that NAD is one of the metabolites of the kynurenine pathway, so if we give NAD (niacin) in enough high doses to psychiatric patients then we can reduce this catabolic pathway and shift it to more serotonin and melatonin synthesis, and the decreased level of neurotoxic metabolites and then treat the patients. Please correct me sir if it is wrong.

    Could you do a follow up of tryptophan in context of Chlostridium Sprogenes with/without prebiotic sugars for healing gut? Seeing this makes me think/worry that boosting tryptophan foods intake without knowing if gut has c. sporogenes might be seriously counter productive.
