Assay of sodium benzoate & caffeine in caffeine injection by simultaneous equation Method-Practical

The video describes the demo of the simultaneous equation method by UV-Visible spectrophotometer for estimating the concentration of the caffeine and sodium benzoate in its injection formulation. Simultaneous equation method of UV spectrophotometric analysis/ Multicomponent analysis. Most of the Pharmaceutical industries manufacture multicomponent formulation, as the treatment of many diseases viz. hypertension, tuberculosis, amoebiasis, bronchitis, etc. requiring symptomatic relief and/or cure. However, only a few of these formulations have an official method of analysis. The spectrophotometric assay of drugs rarely involves the measurement of absorbance of samples containing only one absorbing component.

How to dilute the sample solution if the label claim of both the drug diffrent , like tablet having 2 drugs say paracetamol 325 mg and nimesulide 100 mg ?

Very good question. In such cases u can dilute the formulation at lowest possible concentration which can have notable absorbance. e g. In the example mentioned by u, try to dilute the formulation which will have 2 to 5 mcg/ml concentration of nimesulide and see the results.

sir how much amount of sodium benzoate and caffeine is taken in stock solution i.e. in first 100 ml dilution flask?

In case of sample stock solution as I mentioned we are taking 1 ml of formulation and diluting to 100 ml i.e. in 1st 100 ml dilution flask it contains 1.25 mg/ml of sod. benzoate and 1.25 mg/ml of caffeine.

Because original injection sample concentration is much higher than standard solution, and u know that, Beers lamberts law is obeyed only for dilute solutions, that why we have taken diluted sample.

Usually for exams we give 100 times diluted solution and not the original formulation.
