  • The video describes the problems related to finding out the column efficiency or its performance along with justification. It deals with various formulas to calculate the number of theoretical plates, Height Equivalent to Theoretical Plates (HETP) and resolution of two peaks. In this video, we have taken different types of problems from previous semester papers and discussed the way to solve them.

    120 cm column the sample required 3.8 min to emerge an air bubble required 0.42 min and time required for sample to pass to detector was 0.65 min. Calculate N and HETP ..??
    Sir plz help for solving this problem

    If our resolution is 1.05 and we want 1.5 and column length is 30 then how we can increase resolution by using some different length of column

    • @dr.amolbkhade3003
      @dr.amolbkhade3003  2 ปีที่แล้ว

      If u r using same column packing material with increased length resolution will automatically increase. If u don't want to change length of column u cab decrease SP particle size or use different SP for better resolution.

      For better resolution increase in column length isn't a preferred way to improve it due to retention and selectivity factors are linked with retention volumes which alters the nature of two phases and this also alters the resolution rather do alter the temp of columns which is also a parameter kd depends on

    Sir. 5.54 or 16 in the equation we could see.from where we got those numbers?

      These are the constant values derived mathematically for this equation. Thanks for watching this video.

    Calculate the number of plates for a column having a retention time of 120sec and a base peak width of 22sec? How to solve these problem

    • @dr.amolbkhade3003
      @dr.amolbkhade3003  2 ปีที่แล้ว +1

      I have given the formula in the video i.e. N = 16 x (tR/ tw)2. So in ur question it will be 16x (120/22)2. So it will contains 476 theoretical plates.
