Determining kLa and oxygen transfer rate in bioreactors

  • In aerobic microorganism cultivation processes in bioreactors, it is
necessary to supply nutrients and oxygen to the
microorganism cells. The oxygen supply to the cells is more
problematic because usually, oxygen solubility is
significantly lower compared to glucose and other
components of the nutrient media and therefore kLa, the volumetric oxygen and mass transfer coefficient, and the oxygen transfer rate (OTR) are important characteristics.
kLa shows numerically how efficiently oxygen, which is
introduced through a sparger in the vessel, is dissipated and
distributed in the medium by the mixer. In this short video, I give you a formula to use for small scale bioreactors and three different methods to experimentally determine kLa.

Thanks for doing this! I'm purchasing a bioreactor and as a non-scientist this is very helpful.

@@MarloesPeeters Btw do you have any book recommendations with fundamental knowledge about bioreactors? To basically expand on the lessons from your videos?

@@vincenthus399 : I can recommend this Bioengineering Process Principles book: www.sciencedirect.com/book/9780122208515/bioprocess-engineering-principles (I think you can download a pdf but if not, let me know). I think it is written in a very accessible way.

@@MarloesPeeters Thank you kindly for the recommendation. I could not download the pdf (I think only the first 3 chapters) but I found it somewhere else and reading it now! Many thanks :)

Thank you for your videos. I've watched a couple and these are all high quality and very helpful in preparing for my PhD qualifying exam! Thank you for also citing your sources and tying it to real world examples! It might make your videos too long but plugging in some numbers might help show what an equation means. I had difficulty with some of the stoichiometry because i forgot to convert the qO2 (in mmols) to grams properly.

Thank you, good luck with exam! You are right: practical examples always help and I often see that students confuse units in the exam.

Could u do a vid abt methods of measuring oxygen in bioreactor plz?

    • @MarloesPeeters
      @MarloesPeeters  ปีที่แล้ว +1

Thanks for your response. Just to clarify, do you mean dissolved oxygen concentration? This is normally done with a Clark electrode but there are alternatives

@@MarloesPeeters yes , i mean do concentration...i need the méthodes n techniques of measuring like optical sensors n others...if u know where i can find them plz tell i was looking everywhere but couldn't find it

I only briefly discuss dissolved oxygen concentration in this video: th-cam.com/video/T3DEimHvkAo/w-d-xo.html (around 8:27) where I give two methods - there are other methods though. It does take me a little while to make videos and if you ask me to make a new video, I suggest that you give a small donation via the buymeacoffee link in the description

I fully understand this. Sorry for the late response but if you still need it, I also suggest that you up this paper which discusses the most common sensors/option for DO measurements: doi.org/10.1016/j.talanta.2017.07.088
