I like to flip my agar as soon as it cools so condensation doesnt fall down onto the grow media. Especially with spores, you can always store the cups upsude down and even inoculate them upside down, imo this can help prevent anything falling into the agar when you open the cup. Agar to agar isnt so clean, sometimes the agar will fall off, so I let a2a transfers sit for 48 hours so the transfer has time to bond to the agar then I flip it upside down. Honestly, your videos have gotten me inspired to pick up the hobby again! Thank you for getting me off an 8 year hiatus!

2 months ago I received a back to roots grow kit. Today I have agar plates germinating from cloning, a jar of liquid culture from pink oysters and some rye grain spawn as we speak. Waiting on some spore prints and syringes to arrive now. All this to say thank you and this community! Still learning so so much!

Did my agar ketchup cups following your instructions. Three days later, my agar cups have little white fuzz balls where I swabbed the spores. I have the cups placed in an incubator around 75-80°f . I used a totally sealed airbox and rubbed all the cups and lids and everything I was in contact with in isopropyl alcohol but I did leave lids off while pouring directly i to cups im hoping I didn't contaminate anything but I have little white fuzzy balls growing off the clubs of spores. I also used a normal q-tip that I dipped in alcohol both sides and let dry in my SAB hopefully the alcohol did short work of any bacterial or mild spores.
forgive the noobie question - i come from the world of growing cannabis. you said at 2:54 that those are multiple genetics popping off in the agar. so does that mean like cannabis you can essentially cut out the different "genetics" out of there and put them into different agar cups to separate these genetics - grow them out to find the strongest growth / biggest fruits and rinse/repeat to find the best growth? or is this something that isnt usually done. done a lot of cross breeding cannabis in my 20s in the medial marijuana industry (so legally there YT Lol). appreciate these awesome videos with amazing information. thanks so much.

When you take the spore swab on the mushroom dont you pick up a lot of bacteria along with the spores ? Would uv light help, a little below the the mycelium damaging levels ?

I found some wild beef steak mushrooms, its super rare in my area and I'm hopping to do this with them, once I have some on Agar, are they able to grow in a coir mix? I cant find anyone growing them in a hobby setting.
Of you are using spores from.a print or from a spore syringe they are dirty in and of themselves because mushrooms are fruited in a non sterile emvironment.
I didnt pressure cook my q-tips before taking a spore swab from mushroom gills and storing it. Hopefully itll be alright it makes sense to pressure cook them before touching your spore print again and again but idk
Started on Agar for the first time a few weeks ago. I've done several transfers. To about the third down the line. How do you know when you have a mono culture or whatever? Is the only way to figure it out to grow it out.

Hey man, these vids are great and have inspired me to start my own! I was wondering if you do find some contam in a mason jar should I try and remove it or am I just screw on that jar. I just had to throw out a fully contaminated one with pin mold, I saw it early and just decided to see what would happen (I was lazy and couldn't find if they could be salvaged or not)... the mold was definitely interesting looking haha

what will happen if you inoculate your agar with 2 different types of mushrooms? will the stronger mycelium over take the other one or will you the mycelium mix and you get a different type of mushroom?
My Spores that I dropped on agar just won't grow any mycellium. I don't know why, I did the agar like in many recipes. After that I decided to buy pre made petridishes just to see if I made a mistake with the agar. Still no growth what so ever, I'm really confused

You can, yes, but it's best not to do so much movement over the agar dish and to use a loop or something more suitable for transfering the spores. But it will work! 🙏🍄
I innoculated with syringes and prints onto agar more than a week agao and nothing is growing on it , the temp here is 15-20 *C, any reason why it may not be growing?
You can go from the first transfer if it is viable enough, but it is ideal and standard practice to do a few transfers to clean up the culture because spores are inherently dirty 🙏
Gonna have to do this with my lions mane spores in the martha tent and cover the base with tin foil next time. The black tray i used is covered in white spores im sure itll be a bitch to clean and the racks are rusting from the fogger. Not sure how long i have until they go bad just sitting there though

3 generations of transfers is usually the safest practice, so that you are absolutely sure of your sterile tek.

If I ever get swabs, I plan on clipping the top in order to get at least 6 pieces to put to agar.

They are disposable condiment cups, they don't get pressure cooked, they are good to use right out of the package without sterilizing 🙏
