We have to prepare 100 ppm stock Nitrate solution. Note that 100 pmm is same concentration as 100 ug/mL and 100mg/L. To prepare 100 ppm we have to dissolve 0.7218 gm in 1000 mL VF, if we take 500 mL VF, weight must reduce to half i.e. 0.3609 gm.
To prepare other concentrations, use this simple formula : v1 x s1 = v2 x s2 example : preparation of 10 ppm from 100 ppm solution - v1 is how much volume we have to pipette 100 ppm solutions, we have to find out this, s1 is strength of stock solution which is 100 ppm, v2 is volume of flask we take to prepare 10 ppm solution here it is 100 mL flask, and s2 is strength we are going to make i. e. 10 ppm, From this we know volume we have to pipette becomes 10 mL. That means we have to pipette 10 mL 100 ppm solution and add to 100 ml flask to prepare 10 ppm solution. Exactly similar ways we can prepare calibration standards as we desired.

Measurement would be done at 220nm and 275nm.
Two times minus the conc. At 275 from 220.
Actual absorbance = (Absorbance at 220 nm) - (2 x Absorbance due to 275 nm)
