    안녕하세요! 영상 보다가 궁금한 부분이 있어서 질문 드립니다!
    어떤 분은 centrifuge 돌려서 pellet으로 보관한다고 하셨는데 PBS 있는 채로 -20도 보관해도 되나요?

      안녕하세요, 질문 감사드립니다.
      세포를 수거한 뒤, 바로 lysis를 하지 않고 보관하실 경우 용액 (media, PBS 등)은 최대한 제거하고 pellet만 보관하시는것이 좋습니다.
      PBS에 세포가 들어있는 상태로 보관하게되면 냉각과정에서 얼음결정 등의 요인에 의해 세포가 상당히 많이 파손되고, 다음단계의 진행이 불가능한 상태가 발생하게 됩니다.

      Thanks for the question.
      If the cell types are the same, use distilled water.
      For precise experiments, prepare two beakers, put a solution containing mild detergent in one beaker, and wash there first, and put distilled water in the other beaker and wash the second there.

    안녕하세요. 혹시 트립신에 영향을 받는 세포들을 배양할때 스크랩핑을 통해 세포를 떨어뜨려도 괜찮을까요? 혹여나 물리적으로 손상이 되지 않을까요?

      안녕하세요, 질문 감사드립니다.
      트립신에 민감한 세포의 경우 약간의 비용이 들지만 가장 많이 권하는 방법으로는 TrypLE reagents를 사용하는 것을 권해드립니다. 그나마 손상을 최소화 하면서 늘 쓰던 방법과 동일하게 사용하면 되니까요.
      만약 물리적으로 세포를 떼어내야 한다면 배지 수압을 강하게 하여 쓸어내리는 방법을 사용하는데 회수하는 효율, 그리고 세포의 특성에 따라 회수가 안되는 경우도 있어서 특별하게 권해드릴 만한 방법은 아니라고 판단됩니다.
      수행하시는 연구에서 좋은 결과 나오시기를 기원드립니다.

      A scraper is used when cell lysis solutions can affect the experiment.
      When collecting cells using a scraper, it is easy to check whether the cells are well separated from the bottom of the cell culture plate.

      안녕하세요~ 질문 감사드립니다~ ^^. 실험목적에 따라 trypsin-EDTA처리가 실험에 긍정적 영향을 주지 않거나 다음단계에 영향을 주는 경우에 여러가지 다른 방법으로 세포를 떼어냅니다.
      따라서 scraping도 그러한 목적으로 사용하는 것이기 때문에 trypsin-EDTA를 처리하지 않고 scraping을 실시하시면 됩니다.

      This video is a video that is used in the experimental training of university students.
      And If we were trying to challenge it for multiple plates, definitely on the ice is the best choice. However this is just one plate. So I think we don’t need to perform this one on the ice. It will going to finalize within very short time.
      And also depend on the researchers target, somebody want to analyze about the temperature sensitive protein and/or heat-shock related gene, on the ice is the worst choice. So nobody can telling You must follow this protocol.

      ​ 양예은 To minimize damage to the sample, place the cell culture plate on the ice for the experiment. Temperatures as low as 4 degrees Celsius help to minimize cell destruction and protein damage.

      When experimenting with animal cells taken out of the incubator, there is an advantage that consistent results can be obtained when proceeding on ice.
      However, there are cases where different results can be obtained when conducted on ice in cold shock mechanism studies or active oxygen-related studies.
      This is why it is recommended to proceed according to the circumstances and according to the purpose of the research.
