dear, the dilution factor is considered for final reaction mixture. The reagent is prepared in appropriate concentration priorly. It doesn’t affect your calculation

Because pyrimidine - sugar are stable to acid condition and therefore are not involved in reaction. Purine sugar interaction is comparitively weak therefore seperate out.
