I thought the same thing because that is the one time Herb laughs throughout all the episodes...the camera cuts out after Alan delivers the minefield joke then cuts back to Herb when he's laughing, so ya, I think he genuinely laughed and it was going to be cut out but the producers decided to include the laugh because it was really funny, "and in this business, you never cut funny" (line in My Favorite Year)
when he was on whose line is it anyway and they purposely made each other break character, his real laugh was nothing like this. Just IMO it's all acting here

Derek White: the series jumped the shark at the end of Season 8; Alan was finally growing some balls, and he was developing a slick, deceptive personality; but then of course Season 9 happened, and that plot just went down the memory-hole. If they were SMART, they'd have continued along that line, and had Alan become the main character; then if they wanted Kutcher, he'd come in as their step-brother (since Evelyn was married so many times) and who was also Jake's godfather, and who always stayed away because Charlie used to torture him as a kid. So then Alan becomes the new slacker, and who gets rich by continuing to market cheesy marketing-songs in Charlie's name; and they could continue the "dysfunctional family" angle from there. And if they wanted to make Jake turn stupid, they'd just have him tune out in response to Charlie's death; but he'd be more cynical. Needless to say, Chuck Lorre wouldn't know "funny" if he Big Banged himself in the ass.
Fun fact, Herb’s laugh, that wasn’t in the script, Ryan thought that joke was just that funny and then Marin (Judith) decided to run with it and stay in character. Love when actors and actresses can do so well with improve and mistakes like this.
