When we wash the column *prior* to loading our sample, do we need to wait until all the washing buffer to run through until theres no drop left, or do we load our sample just when the wash buffer meniscus level reaches the matrix?
No no the equilibrtion buffer should pass through the colum entirely. Other wise the column material would not be equilibtayed proberly which might cause poor binding .

It's about 600$ -1000$. Do check out DSHB which is an antibody bank and you can get antibodies at 40$ only
