I feel like Ra's Al Ghul could also work very well as a Justice League threat to the Earth, since the League of Shadows could have their hands in everything and quietly take over the world while the JL has to try and stop them before it's too late.
I heard this pitch on reddit for A Superman movie with Ras, eradicator, and Lex as villains. It worked pretty well, I know it’s not the Justice League, but it was cool.
Lets not forget he was the villain of Tower of Babel and exploited Batman's contingency plans to take down the league. Also I know he doesn't like to use magic but he could be a bigger threat if he starts using it.

I think Larfleeze could work. He could take an interest in Diana’s lasso, Cyborg’s mother box, Aquaman’s trident, etc and come into conflict with the League

Your identity, your essence, your life, your existence. You then becomes a light construct that he can bend to his will. He have no limit to this which is why he have the strength of over 10 thousand, because those are all the people he kill and stole their very being and drain their power and all it to his own. He can also induce an insatiable hunger (greed) within you.

I know a few Wonder Woman villains who are a Justice League level threat they are Grail, Cerce, Maxwell Lord because he was able to mind control Superman, Devastation and Genocide.
Idk WW took out Grail and Circe pretty easily in her recent issue. There is a reason why Tom King is saving Cheetah for this months issue and she wasn’t added in the group effort to take out WW. Cheetah is the dark side of WW in ever regard and if writers give her the power level and respect she deserves she is the strongest WW foe.
Vandal Savage could work well as a Justice League-level threat because he's immortal and inextinguishable. He has plagued the Earth with crime and violence since before the beginning of recorded human history. He is a capable tactician with immense technological prowess and one of DC's most persistent villains, having fought hundreds of heroes throughout history.

I believe the Penguin could have potential to be a Justice league like he could lead a version of the Injustice league and like the Kingpin in the Devil Reign he became the Mayor was able convinced everyone to hate the heroes and have them arrested, why couldn’t they do that when he was mayor and Lex when he was President?

Even though Vandal Savage doesn’t have a normal hero they’re a villain for, I think they’re my favorite “Justice League level threat”. I just love the concept of the Justice League fighting not just a man, but a thousand years of prep time.
I am just imagining Ares LOVING Gorilla Gord or the Yellow/Red Lantern invasions. Making him a secret Final boss for an extended storyline. Also I really liked the Clayface and Poison Ivy analysis. I can already see the potential in both. Poison Ivy just feels fitting being that force of nature Clayface is a mantle or condition that multiple people have taken on. My favorite example is Ethan Benett from The Batman 2003, he starts out as a variation of 2 Face, a personal villain for Bruce, Joker and Gotham PD, and eventually gets reformed, but his legacy continued into a stronger and more malicious person. The next generation of Clayface being an out of work actor. So the idea of a few more generations of Clayfaces going insane and growing more powerful and toxic like a virus culminating into a Justice League level threat because it snowballed over time could be a really great threat. Because someone would fully embrace the monster they've turned into because they likely sought it out or wished for it to be.

I don’t really like Poison Ivy as an evil villain, especially since humans are part of nature, too. So I’d propose smart Bane, or like you said, empowered Clayface. Also, I think Atrocitus is more interesting than Sinestro, especially since we could change how the red rings work slightly, and have it be so Atrocitus (or really anyone) can FORCE a red ring onto someone, and then it’s still one of those things where they die if it’s removed. Because I would LOVE to see Bruce/Batman consumed by rage, and STOPPED by Jason. Basically, a reverse of Under The Red Hood. Where Jason kills Bruce… and then probably mutters something like, “Sorry… dad…” and boom, right in the feels. Everyone loves it. 🎉
Okay mantle until he becomes Batman or Nightwing. But personally I prefer he gets something more unique motif. Like the Crow thing like in his manga. Also we already got two robins with red in the name 😅.

I 100% agree with Gorilla Grodd as the choice for Flash like if the other speedsters wanted to be they could be rivals to the Justice League like with most of Superman villains but it’s more interesting to not have that. The other option I think is just the Captain Cold and his gang as a group amped and the Justice League having to deal with them because the Flash alone can’t stop them for whatever reason. I think any of Wonder Woman’s big league villains could be thrown in as a JL enemy because like Superman she is really powerful and they have something backing them so they can pull in followers to stall and what not and can be given motivations to go after the whole lot neatly. Batman I honestly think none of his villains really fill the role. Not even Ra’s Al Ghul. Ra’s is just using Batman’s plans and isn’t really a foil to the JL but used as a way to explore the trust and relationship between them. The enemy is more Batman’s mistrust than Ra’s and for Poison Ivy there’s just more interesting things for her to go against when hyper powered then JL such as swamp thing For me with Green Lantern like any of the cores could be used but they suck as an answer for a similar reason to Batman’s villains they just are better as a contained story to themselves or against different villains to the justice league. Honestly think Larfleeze is the best of them cause at least he’s very different comparably. But he’s also not really main event Star and more an obstacle that you have to manoeuvre to get something to beat the real foe. For Superman Lex is cool but he’s cooler against either Superman or Batman alone without the Justice League and Ra’s or Vandal Savage are probably better anti-Justice League group leaders. I think Doomsday sucks cause he fills the same niche as every other Justice League villain like Anti-Monitor or Darkseid and has no real motivations. I’m going to say Toyman could be done more interestingly against the Justice League or Zod can be done very boringly but is better than Doomsday still imo. But the two real best are Mr. Myztplk and Batmite I mean come on. Clearly they need the whole Justice League and more to be in on getting rid of them.
