#Retrieving_sequence_in_correct_orientation - In this lecture, I describe the basic concept of Sequence - #Plus #strand, #minus #strand, #complement and #reverse #complement and also explain the steps to #retrieve the sequence of the gene from #UCSC #genome #Browser

Respected sir
I am a biotechnology student and being a biotechnologist i personally feel that a biotechnologist must be expert in bioinformatics tools.
I have knowledge of wet lab but not of dry lab
I have taking the lectures from your lectures series and you have taught in very good way.
Thanks you and keep it up
Thanks for sharing, professor.
My pleasure! Do share with your peer group too.
Very nicely explained Sir. If we do not mask the repeats then also it will be shown in the DNA sequence ?
Yess! Then it will be shown as a regular sequence I.e. you won’t be able to discriminate between what’s a repeat sequence and what’s not !
Thank you, Sir, for your clear explanation of the reverse complementation. But I am confused. How do we determine a DNA strand as either + or - (plus or negative) strand? What is the reference to it?
For this you have to understand how a DNA is generally sequenced - Most Sequencing technologies use DNA polymerase and Forward and reverse primers to sequence the fragment from both ends - the sequence coming from the forward primer is considered plus - check out my Sanger sequencing video in the channel also NGS play list
why there are lower case letters?
You may have selected for repeat masking to lower case option in the get dna window - UCSC browser
