  • The binary vector is a two plasmid vector system created by modifying the Ti plasmid from Agrobacterium. One of the plasmid in the pair carries DNA insert and other carries the vir genes for the transfer of the insert from Agrobacterium to plants.     Good explanation overall but I have a question. The helper plasmid only contains Vir gene then how does kanR develop in it. And how it also grows on media containing kanamycin?

    Extremely helping thank you. Question - Is this process only specific to neomycin-resistant gene transfer or can it be the same for inducing other crop resistances, ie insect pest resistance using the Bt protein into maize crop? Is the process very different then?

      Yes, the Ti plasmid vector systems can be used to induce insect resistance, conditional resistance, and metabolite production in plants. The process remains same, only the gene of interest changes.

    what if plant specific selectable marker in a Ti plasmid binary vector has been disrupted during the recombinant DNA formation with a specific DNA insert .whether this Ti-plasmid Still be used for plant transformation ?

      Hello Somya,
      So the two plasmids do not undergo recombination event. They stay as individual entities. If you're talking about the construction of the vector and adding various sequences in continuity, then that is taken care by sites of homology and complementary base pairs. Also, use of linkers and adaptors can help in joining two sequences.
