Let's not forget SPN referenced itself unknowingly when Sam is reading a spell in S3 to keep Bella from drowning via ghost. He actually calls upon "Castiel". And this is before Cas is even a consideration on the show.
But if Supernatural the show exists in Legends Of Tomorrow, then wouldn't Sara recognize Ruby's first actress as her sister Laurel and Jack's mom as Ray's wife Nora?
That last one is less of a Supernatural reference per se and more of a continuing tradition because I think he also played a character named Bobby Singer on "Dead Wood". It's kind of a running gag. There was also a SyFy Channel series called Superstition that was basically a race swapped Supernatural except it was a father and son and their family in the first episode the father and son are going after weapons the father keeps in the trunk of his car and the son says "arsenal in the trunk like Sam and Dean?" It only ran for one season and while the premise was kind of familiar I think it was a fairly good show.
I'm pretty sure that in 15x03 of Supernatural, Castiel had to do a chant and then a demon possessing Jack's body told him had to sing it, but we unfortunately never saw him sing it bc it cut to another scene
