    Mam is this method CTAB used for DNA extraction from plant materials or only for plant cells. I am confused. 😑

    Moreover, Can we use ice-chilled ctab buffer instead of liquid nitrogen ?
    After adding eq. Vol of chl. and IAA, we get to centrifuge the sample, kindly mention the rpm and time.

      Don't use chilled CTAB, for CTAB to be activated u need to preheat the extraction buffer @65.C for 15 min. So if u don't have liquid nitrogen u can directly add the pre heated extraction buffer to ur sample. May I ask what sample are u using?

      U can do the Centrifuge@13000rpm for 1-2 min.

    Make a video on dna extraction from whole blood or plasma. Secondly, i want to know the full recipe of ctab extraction buffer considering i am short with pvp. That would be an honour.

      Well we don't use much amount of PVP but if u are short of it u can treat the sample with chloroform and isoamyl alcohol two times and carefully separate the uppermost layer

    So even for seed will we follow the same method? Actually dna isolation is not a routine work in our lab so i was looking for a method for plant dna isolation for quite sometime now.

      For seed also you need to crush open the seed for isolating a DNA... The procedure is basically same. Just take appropriate precautions at each step. All the best

    What is main difference in isolation of DNA from bacterial cells and from plant m cnfuse dua to lot of chemicals and steps

      Since the cell wall of bacteria is not as complicated as plant u can easily use some chemical, or enzymatic methods for its lysis but because plants have much complicated cell wall different chemicals are used along with mechanical methods and certain chemicals are used to neutralize the metabolites released during crushing of plant tissue. So the chemical utilization is different in both otherwise the basic concept is very similar

      This is the basic procedure, u can use it... But recently other methods have been developed which are more significant and improves the quality of genome sequencing

    how to dissolve 5M NaCl in 500ml dwater

      To make 500ml of 5M NaCl solution use the formula--> gm of NaCl = (mol. Wt of NaCl * Molarity* vol. To make) /1000
      =( 58.44*5M *500ml) /1000 = 146.1gm
      Dissolve 146.1 gm NaCl in 500ml D/w
