OK, so if a drug is more protein bound, it will tend to pass more slowly into other compartments. Permeability of barriers also depends on pH of the compartment. Higher binding capacity means less movement into other compartments. I think I get it. Thanks for the overview! Oh, and it looks like you spelled permeability wrong. :-P

Does anybody know why the conc. drug in plasma is the only one discussed? Is that a standardized form of measurement for determining the effect of a drug? If so, are there any variables that might be overlooked, such as if a specific patient has abnormal permeability been one compartment and another? e.g. For a drug that's active in the CNF, and the patient has higher permeability between intracellular fluid and extracellular fluid?

Although somewhat late... the Vd is = Dose/(Cp * kg). Dose is in mg and Cp is the peak concentration of the drug (mg/L) times the weight of the person (kg). So you have mg/(mg/L)(kg) or mg * L/mg * kg so mg cancels out and Vd is in L/mg units.
***** How did you get 15/10 mg/kg? You just start with 5 x 3/70 = 15/70. For the units, it's L/kg x mg/L, so you just take the "L" out, which leaves you with mg/kg. What does it mean 3 by 70th ? Does it indicate that the plasma concentration ( known concentration ) is 3*70 = 210mg /liter ?
