A few days before we did this experiment in the lab but didn't got this level of clearity. It's just amazing to do this by your own hands, Great job sir🥳🥳🥳
Thank you! We just did this experiment a couple of days ago, but I didn't have time to sketch a couple of the mitotic phases. And anaphase happens so quickly that none of our fixed cells were in it (the best was early telophase). Really great video for filling in my student logbook (obviously I'll note which sketches come from this video & which I saw in person).
Excellent explanation of how to prepare the material, and the photos at the end showing stages of mitosis are among the only pictures I’ve seen that actually look like the textbook diagrams say they should - many thanks!!
Thank you. Hope it helps. Dont hesitate to ask any questions from this. Will also be jotting down a brief theoretical description of mitosis is the vdo description later.
My board exam done I did same like you did but mam said I will see it under microscope you leave hope she will be happy 😊 my also same cloudy and colorless thing was made just like you
Hi. The same setup will work for all phones. Alternatively, you can invest some money and buy a phone to microscope adapter device, you can find lots of cheap priced options on amazon and other websites.
Thank you so much for this video Actually we were told in our school that the cells taught by you... are chromatids and on the basis of their concentration they were telling us about anaphase and metaphase🙄😑
Q. What is the requirement of squashing the root tip? Q. Any other substitute for acetocarmine ? Q. What is the reason behind heating the root tip with n/10 hcl and acetorcarmine ?
1. Specimen to be observed under ordinary light microscope should be transparent to near transparent which means specimen has to be sufficiently spread in a thin layer. If you observe the root without squashing, you will see nothing but a giant dark blob that's out of focus most of the time. 2. Aceto-orcein, Giemsa. Some even use methylene blue although i have never tried that. 3. heating speeds up the cell wall plasmodesmatal softening and disintegrating process by the HCl so that you achieve a thin layer squash easily.
No problem. But there is a paper that mentions the negative impact that prolonged HCl hydrolysis can have on the staining power of acetocarmine/orcein stain. So if u r in a hurry, its allright take the shortcut. Otherwise, better do the hydrolysis and staining steps separately.
4:59 WOW!!!!! This is probably the BEST REAL PICTURE OF ANAPHASE!!!!!
