This guy works really well, he straight and to the point and he only takes 9-10 mins to explain things where as in class the instructor takes 40-50 mins of just throwing info at you and you don't really get anything out it. Really I appreciate your videos you obviously know your materials.

In contrast, the CAP-cAMP system is an example of positive control, because expression of the lac operon requires the presence of an activating signal-in this case, the interaction of the CAP-cAMP complex with the CAP region.

Both lac operon and trp operon are negative control operons because repressors are involved. However, the lac operon is inducible (induced by allolactose) whilst the trp operon is repressible (corepressor tryptophan).
Dude I live in India and in my high school i almost spent a day or 2 just trying to figure out this operon..reading tens of books but still couldn't understand much. Dr. Bozeman, you might have it backwards at 6:39 According to Wikipedia lac operon is Negative Inducible -- negative because in it's normal state a Repressor is needed to turn it OFF--and Inducible because expression can be turned back on when induced (allolactose). "The lac operon is a negatively controlled inducible operon, where the inducer molecule is allolactose"

Thanks for the info. But i don't think positive regulation is the correct terminology to use for the allolactose binding to the lac repressor. Positive regulation is specifically the binding of an activator to it's regulatory sequence and thereby increasing gene product.

Great video for clarifying some major aspects of gene regulation. It is almost important to take into consideration the physical state of the chromatin involved in the process for this is another major factor which impacts gene expression.

I just learned in Microbiology class that the lac Operon is both Negative and Positive Regulation. It is negative due to the repressor being bound to the operator by default. BUT when glucose levels are low, cAMP comes along and binds to CAP, this allows CAP to bind to the Promoter site and allow RNA Polymerase to bind to the Promoter and synthesize B-Galactosidase; all of this can only happen if the CAP is on and the repressor is off. The repressor comes off in the pressence of allilactose.

However, he made a couple of correction from your video, so hopefully it will be helpful for everyone and editor. "making genes" is said a few times in the video when instead it should be "making RNA". Lastly, the mRNA made from the bacterial lac operon is one polycistronic mRNA encoding three separate proteins. It is stated incorrectly in the video that three separate mRNAs are made from the lac operon.

Campbell biology book 10th edition says"regulation of both the trp and lac operons are switched off by the activate form of the repressor protein. " but you may feel that lactose binded repressor make the transcription work. But remember that activated repressor means it can bind to the promoter .so lac repressor is activated as the moment it is made. That means ,it's intact form is activated. But the way that you may think , i mean the allolactose (the isomer of lactose formed in Small amount which actually bind with the repressor) make the gene active is not totally wrong. That's why Campbell says trp operon is a repressible operon and lac operon as inducible operon.**(Campbell biology 10th edition page 418 419 420/ unit 18)Both operons are pertaining to negative gene regulation and example for positive regulation is catabolite activator protein (cap regulation). Bozeman is not wrong and the case is about terminology. The (positive) must be corrected as inducible and negative must be corrected as repressible.☺

I missed some lessons on this but understood everything perfectly thanks to this video! A lot of videos are only good for revision purposes so you only understand them once you've already been taught the topic, but this video was excellent for someone like me who hadn't visited the topic at all.
