    Not a good idea to test continuity or resistance on a powered circuit. It's a bad habit that could hurt you in certain contexts if you don't make it part of your muscle memory. Disconnect the battery, first, or better: measure the Voltage between the two points. It'll be 0v if there's continuity, higher if it's broken

    does it matter where the black/ red leads go on the fuses?

      No it doesn't matter. This is a continuity test. So as long as there's continuity, meaning the fuse is OK. That's all you want.

    Does the ignition have to be ON inorder to test certain fuses?

      No, fuses can always be tested without power from the car. If the ignition is on then power is running through them and it’s more dangerous to test the fuses so I would recommend against it.

    What if I want to test a fuse to see if current is flowing through when the car is on or off

      You would use a test light between the two points. If the light is on then current is flowing through the fuse
