How to Make Agar Nutrient Media for Orchid Seeds - Quick and Easy Process

- In today's episode, I'm sharing how to make the agar media for growing orchid seeds. I'll share my process on how to create the nutrient solution for sowing orchid seeds. This is the germination media used in aseptic orchid seed sowing. It's the first step of several to grow orchids from seed. There are many ways to do this and many variations as well...this is what I do and what works for me. I'll also talk about the reasoning behind this procedure and why I do what I do.
I'm using Phytotech P723 Orchid Sowing Media for epiphytic orchids. Terrestrial orchids require a different recipe for agar media. For sterilization, I steamed the solution for 40 minutes. If you were to autoclave it or use a pressure cooker (121C at 15psi) with the same volume (50ml), then it needs about 15-20 minutes.

Thanks for the update 😊🙏 really excited about where this is going. Great explanation and really good tips. Fully agree with you on starting with the basics and getting that right. The commercial guys have it so easy with their laminar flow and autoclaves 😅. I keep on reminding myself that these techniques were around way before any of the fancy equipment.
Haha yea! Fancy equipment definitely helps speed up the task, but having good technique is better still. A lot can be done with simple items, so I don’t worry about having the “right” things. 😁

Hey I know I am kinda late to the party but I really need to know if its possible to just boil/cook the jars/flasks? I dont have a pressure cooker but I have managed to polinate a orchid of mine and really want to continue the work.

Hey Jess! Thanks for making this video. I've been watching some mushroom cultivation videos and I wonder if I can use mason jars with self-healing injection ports to cut my risk of contamination? Let me know what you think! I think I can instantpot-sterilize my agar solution in a self-healing mason jar, then inject the dry seed + peroxide solution via the self-healing port. Do you know which gauge needles would be large enough for orchid seed?
It might work, I haven’t quite considered it. Some orchid seeds are small, about the size of mushroom spores so those might work. For larger seeds like from complex Dendrobiums or other types, they might be too large for a typical syringe. I had trouble getting some seed out with a plastic 2ml syringe with no needle tip…really just depends on the size of the seed
The best way to cut on contamination is to use green seedpods, they come sterile from nature. I flame a lancet, divide the seed-pot and use a sterile tool to pour few seeds in the flask.
Peroxide, syringes, Clorox... are noise in the system if you have access to a laminar flow hood.

after you planted the seeds whereby you incubate the jar? and for long?
After seeding the jars, I leave it alone in a slightly lighted location. It took about 3 weeks for strong germination and it still continues to grow in flask. I am waiting for the first leaves and roots to grow before I think about reflasking into a larger space. I believe the plants stay in flask anywhere from 1-2 years.
If you are doing solid media, there is a germination media and a replate media. The germination depends on the species of orchid. Some take a few weeks, some take much longer. The species and the medium you are using makes the difference. I like to move my protocorns to my replated media and let them to growth there. Sometimes you have to replate your replate if all your protocorns made it to orchids.

I sometimes get plants (seedlings) that could need some more tlc would agar be a good idea to grow them on ?
If the plants are small enough, it may benefit from a little more time in agar. It would need more nutrients than the P723 media since that is usually used to germinate seeds. P748 or something similar (replate or maintenance media) would be better since it has more nutrients to sustain a larger plant.

You can search peer review papers. There are some specially in Brazil and Peru using krystalon orange fertilizer, sugar and cooking agar and seems to work fine.

The media is put away for 3 days to check for contamination. If it does not grow mold or bacteria, it’s good to use. Not for outside.
