Orchid Flasking, Breeding & Propagation
This is a detailed walk through of how I sow dry orchid seeds using hydrogen peroxide to sterilize / disinfect the seeds. I use a glovebox NOT a laminar flow hood (because it's cheaper and saves space in my small apartment). In this video tutorial, I'm using a seed pod of Phalaenopsis pallens x self, which I sowed myself in January of 2019 (6 months ago).

Justin, great video! You explain things very thoroughly and clearly. Another benefit of using dry seed is that there is very low risk of transmitting virus. A virused plant can be used as a pod parent (this is sometimes done with rare, heirloom but virused plants - using a virused plant as a pollen donor will infect the pod plant so is not recommended) and the seed will be virus-free - but cutting open a green pod can transfer virus from the pod to the seed via the sap in the pod wall. With dry seed, the pod dries and then dehisces, and the seed do not come into contact with any sap. I staple a paper coffee filter around a pod as soon as it starts to yellow, to prevent seed blowing away when the pod splits. Dry seed can also be stored but they must be dry or they will get killed by mold - there are chemical ways of drying them (complicated) or you can bake some raw rice and then put the seed in an open dish on top of the cooled rice in an airtight container and the rice should absorb any moister - then fold the seed in waxed paper, label, and store in a sealed jar - one of those desiccant pouches from a bottle of aspirin or some food products can be thrown in to keep it nice and dry - and refrigerate. I'm told the seed can keep a few years, depending on species. PS - your kitchen is spotless compared to mine! I find it ironic that I can maintain sterile conditions for sowing and re-plating but can't seem to mop my kitchen floor, LOL.

This is so cool man! I work in an orchid propagation lab but it’s awesome to see the process being done at home. This video will come in handy whenever I try this at home
I'm a conservation biology student and..I don't want to get too detailed here, but I need to try to clone a leafless species from the root tip and or from the spike. I can probably use a laminar flow hood but I've got no experience with micropropagation... if I sent you a piece of root, could you sterilize it and toss it in a flask? It doesn't have an active root tip. I don't know if that matters. I don't even know if this piece of root will survive after the holiday, but I will also have a spike in the future if I manage to not kill the plant this semester, and I would like to try cloning it or having someone try to clone it for me. A friend of mine has lindenii and I keep telling this person that attempts at cloning it from the nodes on an inflorescence would be a worthwhile project. There is a video about how to clone orchids from spikes by orchidspecies but he doesn't sell the kits anymore. I don't know what the medium he uses is or if it would work for the species I want to clone. I do know what medium works well for growing this species from seed, so perhaps replating onto that after a plantlet forms would work.. anyway, I would really like to talk to you more about this in-depth. If you sell any little cloning kits like orchidspecies used to, that would be really handy.. if not, if you could help me choose a medium that sounds similar to what orchidspecies use or if your lab uses one for this kind of thing, I would appreciate it so much and if I am successfull, I wouldn't mind sharing one of these orchids, and I have a sneaking suspicion it is a species you would very much like to have.

Yeah, wow and woah, man. I’ve rescued one grown plant from a co-worker and root shocked/killed a second through ignorance. So for fun wondered how to germinate and found your video early on in the search. What a world of detail and precision I am seeing this is! Thank you that was VERY informative! I had seedpods in the past, as I pollinated just for fun but didn't know what to do with the seeds then. After seeing this, I'm 100% certain, I will never attempt to seed grow orchids. At this point I'll leave it to the experts and just buy the finished plants.

This is an interesting subject. I didn't realize the care it takes to grow orchids from seed. I bought orchid seeds recently and coming across your video has given me instruction I didn't know I needed.
Sometimes I buy seeds online, they are scammers, they sent my aunt crushed seeds... how did they come out?
Very informative and pretty indicative of why I can't imagine doing my own flasking, I'm a klutz and scatterbrained, contamination would be inevitable. I just let a bunch of seed go in the wind recently, maybe they will find a place to germinate. I probably should send the pods on my Den Samurai and Den antennatum D'albersti (ants keep pollinating them!) to someone, I don't need them but maybe someone else can make money off them.
haha - so funny. Yeah, I get that side of it...I'm pretty "relaxed' with the whole process myself. I just make sure I write in the methods that allow me to shortcut success...which doesn't even sound like english. TL;DR - I make it as easy as possible. Hahah

Hi there, I wanted to suggest the use of micropore tape (3 um pores, 3M brand for ex.) to seal the medium containers. I have been working in plant biology labs for a long time and breathable tape is standard practice when cultivating plants on agar. I know some people use a wax based seal however the lack of air exchange inhibits plant growth (at least for rapidly growing plants). It's pretty cheap and might reduce the number of transfers and maybe even required growth time, who knows!
Good video as usual. Would like to see the project all the way through to the plant grows up at least 6 months old. Many people on TH-cam do these videos greatest best way to do something - and then we never see if their way actually works or if the operation was a success but the patient died type of situation. For all we know the dog they coated in some magic mystery potion for ticks died or the fumes drove the people out of the house or they got some allergy from it.
I would suggest using horticultural/agricultural surfactant that doesn’t have fragrance, oxidizers, brighteners, sodium x, and the dozens of other chemicals in it. And use a mask -we all have them readily a available these days - your mouth has lots of bacteria etc. I also believe in first washing with an antibacterial soap to remove “gross” visible soap then use Physan 20 mixed to the correct dosage as they recommend under the Application tab and go to Orchids. You want to dip and splash your clean hands around, let it dry, and then keep your hands wet with the solution the whole time you are dealing with the plants. I make up a fresh 3 gal covered bucket ever couple of days Spray down your surfaces in between plants and use plain newsprint paper to contain all the old mix, dirty an old parts cut off the plants, etc, roll it up and put in trash. I also keep a large bucket of soapy, bleachy water made up to initially drop dirty pots into. Later, I wash them as clean as I can get them and rinse, then drop them in a soak bucket of Physan20 then set them in my hot sunny south west full sun deck for several weeks. Sunlight is an awesome sanitizer clay pots. Physan20. In fact, with questionable clay pots that have gone through everything from these to baking in the oven - I will leave them sitting in the deck railings for 6 months or so. For those who remember Bob Scully, he felt the best way to do clay pots was to refire in a kiln as that got rid of anything from simple dirt to viruses (2000F). There is an article on St Augustine Orchid Society that that gives the procedure for cleaning, disinfecting and sanitizing clay pots including neutralizing the mineral deposits. I use Physan to generally clean tools but use a dip of 91% alcohol as the less water in the alcohol, less rust and then while wet I burn the blades. Physan20 is a bactericide, algaecide, fungicide, viruscide. I use it to sterilize seeds, to wash agar off the seedlings when unflasking, to soak pots, to treat plants when repotting whether diseased or not, water the medium down with Physan 20. Their website physan.com under Orchids lists directs and dose strength and time for several nasty diseases including Erwinia. Also mix to dosage requirement in gallon jug, put in pump spray bottle and spray plants down once every 7-14 days to help prevent disease especially in the crowns of phals, Vanda.
Surfactant: Anyone who applies fungicides or herbicides in your garden or on Orchids knows many require non ionic surfactant to allow the product to adhere to the leaf surface so it stays long enough to work properly. Also read your labels for each brand of product as the active ingredient may be the same, but the strength and proprietary ingredients may be different and have varying instructions as to dosage, usage, plants tolerable and those sensitive to the product. There are a number of good non ionic surfactants - One I use is Hi Yield as a surfactant - it’s cheap and does the job. Just make sure you check what type of surfactant is required with the product and with what type of plants - vegetable crops is usually different than say ornamentals. If there is a range - I use the lowest. Id I have questions, the manufacturers all have customer service telephone numbers and email addresses. I find them all very friend,y. Any local farm store will have these types of products (including the herbicides, fungicides, etc) cheaper than like online or Amazon. If they don’t carry it, many will order it for you. I’m finding more and more we are paying way too much sometimes double for online items.
I also like using jars that will stand up to boiling in a canning pot (canning jars - they come in sizes from 4 Oz to gallon). Food containers should be boil resistant also but be careful with cosmetics, etc type jars that might not be sanitizer temp and will burst when doing this. Also, if you start your sanitization and cleaning process in your dishwasher, make sure your dishwasher undergoes thorough cleaning process. There is a lot of gunk that sits in the bottom and stuff splashes back on the dishes. It’s pretty gross and most people don’t realize. It’s the same for washing machines - after seeing one taken apart, I now use Afresh once a month. Clothes smell a lot better now too. You need to realize vinegar and baking soda are recognized as good cleaners but aren’t approved by the EPA as disinfectants or sanitizers. The EPA gives a list of active ingredients for you to look for in the brands you like which are strong enough to do the job. For plants check the specific fungal or bacteria to make sure the active ingredient will kill such and such. Some pathogens protect themselves in such a way, that they may not be affected by something that kills everything else. Many items like bleach must be kept soaking wet and in contact for a specified 10-20 minutes depending on what the pathogen. EPA lists household cleaning vinegar which is white Vinegar as only listed strong enough for 3 specific pathogens I believe. Do not use other forms of household like ACV, wine, rice, etc vinegars. And don’t mix vinegar with ammonia, bleach, hydrogen peroxide or products with those in the ingredient list.
And it’s not always the shipping because I have had exact products shipped from one vendor for $7-9 dollars and the next vendor wants $50-60. Crazy and a ripoff. They always blame it on the shipper but that just isn’t the case. I recently shipped something - at UPS they wanted $17 for packaging plus $47 to ship. I said I’d have to check with post office - they said I can check for you and told me it would be $60. I said I would go home and find packing myself and come back. Stopped at post office, they packed it for free, and it cost me $9.90, got there in 3 days including the day I shipped. I won’t be using that UPS store ever again. I just had one said it would cost $60 to ship 15 little net pots and someone else it was under $10. Both said post office. Had one that charged me a fortune for bark I really needed said it was going UPS. I paid, they shipped, got it in 5 days and it came by the post office. I took it back to the post office to find out how much it actually cost and there was a $40 dollar difference - contacted the vendor and asked for a refund. They were oh really, don’t know how that could happen, bla-bla - right. They know exactly what it costs and who it gets shipped - some of these are making a big profit. They are using a system that figures at such and such rate, you pay it, but they actually ship the cheapest way postal and you get taken to the cleaners. It’s not right. Sorry - got off on soap box.

A single orchid may produce millions of seeds but one a few will take hold and become mature plants. In captivity we want to maximize the success rate.

A traditional still air box with large open arm holes does a better job of keeping things sterile than trying to imitate a glove box. The seal around the gloves just moves around too much air while you're working which defeats the purpose. Real glove boxes are used more for working with hazardous materials or things that need to be handled under certain gasses, not for sterile work. This is well documented in the mycology hobby and you can find a wealth of info on the shroomery.

I hope you give it a shot-while it can be a lot of work to get started, the reward is pretty gratifying. For example, one of my Phragmipedium seedlings is variegated! I’ve never seen any phrags like it - but it’s still so small. Haha

as a veterinary healthcare worker with experience in surgery... this is so overboard.... like lab animal innoculation style overboard.... which is kinda perfect for athome hobbiest work (from the contamination to aseptic technique compensations... it seems just a stronger amount of contactlessness is required :) ) 🥺
I have experience in aseptic technique and innoculation and such, so when it comes to some of the materials i'm probably going to change a few things (ex: syringe with caps, test tubes for higher isolation rather than jars, box hood rather than glove box because with high enough aseptic technique, that's all that in theory shouuuuld be necissary (again, in theory), covid style surgical mask, maybe nitrile gloves instead of the glove box.... but beyond that... i'm so heavily impressed with the aseptic technique he shows and this video gives me such a strong direction in how to start growing orchids from seed!!! Like seriously, the way he uses hydrogen peroxide is just... brilliant!!! I've got to bring that up with my old professors to see what they think. :D Thank you!!!

What's you success rate for the germination? Will you do a replate video in the future. Quite curious on how you can prevent contamination then.
Success rate? No idea on exact numbers, mainly because I don't have insight into how many individual seeds are sown. With the phrags...germination was generally lower...but that could be from hybrid viability, from keeping them in peroxide for too long, or from them not liking the media...I really don't have a good way of extracting precise answers for that.
With regards to replating...it's mostly the same process in the glove box; but I am still learning that. I did my first "real" replate last week...and before that, a bunch of spreads. Spreading protocorms is a lot easier than replating seedlings with roots. I will do the replate video one day...but for now, I need to understand the process better before I feel confident making a video about it.
Think of using Hibaclense... It's surgical soap and wont be as harsh on your hands as bleach water. Very good video.

I enjoyed this video! Great tips for use of hydrogen peroxide. I will try that next time I sow seed. I used a glove box for many years. Recently bought a HEPA filter and built a homemade flow-hood around it. Works good so far.

I have a question regarding the pressure cooker you use for sterilizing. You mentioned that it must be 15 PSI. In the UK, I've only found a pressure cooker that can pressurize up to 100 kPa/14.5 PSI. Would this be sufficient, or does it specifically need to be 15 PSI/121 Celsius?

Use alcohol with friction on your hands (wash with soap and dry before for extra percautions) not bleach. No paper towel drying necessary. Bleach kill time is 5mins generally so unless it sits on the surface for the stated kill time it's not as effective.

You could buy some surgical desinfectant for your hands. It would be much more gentle for your skin.

G'day from Perth Australia, Ive been enjoying your posts for a while, thank you. I think I have had one of my Paphs pollinated and I think it is growing a seed pod?? I wish I could show you a picture of it, but, from the old flower spike just days after the flower fell, the spike has become thicker and ridged, somewhat resembling a yellow long necked squash/pumpkin ( obviously not that big) What do you think?
I’ve been keeping plants for a while, and I pollinated two of my orchids (a brassocattleya and an oncidium gold dust). I am planning on flasking them and am wondering if the green pod method is better for a first time, or using a split pod. The oncidium seed pods are relatively small, so I’m wondering if that would make it more difficult to do green flasking. Also when you take the jars out of the pressure cooker, do you just wait till it’s fully cooled and then open it and tighten the jars by hand and seal them? Just curious how you prevent contamination when making the agar. Also do you find more contamination with the green pod method or the method you show in this video?

I would be very interested in seeing a side-by-side of this method vs just sterilizing outside of the pod w/ Isopropyl. I feel like you would end up with similar contam rates.
I would think that the interior of the pod is already sterile. So the whole syringe deal seems like a lot of unnecessary work.
Aaron Wanker - depends, right? Example...I once tried the green pod method with a Phrag seed pod-the disinfectant got into the seed pod and rendered all the seeds useless. Another time I dropped the seedpod in the glovebox and ended up with all contaminated flasks.
Other times, I have sowed dry seeds without sterilizing them at all-a pod that split unexpectedly and had very few seeds (and therefore the hydrogen peroxide would not have worked). I lost 1 of 3 flasks to contamination, but still have 2 with seedlings and no contam.
I think you should use whichever method appeals to you...for me, a couple drops of H2O2 from a syringe into a flask is much faster and easier than fumbling with tweezers and a slipper seed pod in a glovebox with reduced visibility-and if there are any screwups with dry seed, I have more to try again. If you screw up a green pod...it’s done.
It’s a Cost-benefit and you need to decide what works best for you. There are lots of tutorials on greenpod sowing, but not so many on dry seed.

How on earth can orchid ever spread by seed in nature?? Is my biggest question after watching this.
In nature they grow with symbiotic fungi and of course conditions in Asia/south America/Africa are more consistent so they never go too dry at the early stages of life. We do it this way because it’s much easier than trying to grow the fungi

Love your videos. But I am Confused as to where I get the mother jar from in the first place. Where can you buy it or what is it? Or can you buy a mother flask with orchid seedlings growing in it already? As I am in ireland? And you can not buy anything like this here. There are no orchid shops to even find moss to buy is impossible. Even to buy an orchid plant costs an arm & a leg 🤪

Mmmmm - Ryan, I'm not sure that one seed would be possible? Orchid seed is fine like dust - have you ordered something online and this is why you think you have one seed?

It’s called Agar - you can buy the powder online and add distilled water n boil it together and add to sterilized flasks
Is there is there a way u can send your orchid seeds to a lab so they can germinate and then they give u your seedling in a few years?

A suggestion do not use your finger when you are shaken your solution with the seeds, you should put the needle back in the syringe, la solution won't not be lost because the needle has his own cover and will not be loosed, I am telling you because I am diabetic and I use insulin every day, in this way you will not contaminated the solution using your finger, after the 10 minutes, you can pull out the needle and your solution will be perfect, believe me. Hi Roberto, Tnx for the comment and thoughts. The needles I have do not have an airtight seal, and when you mix the seeds with peroxide it fizzes, creating gas and pushing fluid out. You need to use your finger so you create a tight seal allowing you to make hard compressions and make a vacuum by pushing and pulling the plunger while the tip is plugged-you need to do this to ensure the peroxide gets into any areas where air bubbles are. I have found as long as you wash your hands with soap and bleach prior to, contamination shouldn’t be much of an issue.
I love the info provided in the video, but do have a couple questions regarding the process involving the pressure cooker. When you put your jars in your pressure cooker, do you stack them? Can you stack them? Are they sitting in water? Does it matter how much water you put in the pressure cooker?
Good questions! Answers: When I put the jars in the pressure cooker, the lids are only resting on top very loosely...not tight; I want the air to pass in and out during pressure cooking and cooldown, then I'll close the lid. This prevents an air "suck" when you open the flask at time of flasking...but it means you have to let the whole pressure cooker fully cool, before you open it.
2) Yes you can stack them, and I do
3) My pressure cooker has a canning "lift" that sets the jars above the water level. I don't think it's a major problem if water touches the jars, but the way my pressure cooker is setup, they don't touch the water.
4) You need to use at least enough water to evaporate to make pressure, and then also account for water cast off from the top during the cook. I use about 4 glasses of water...or 48oz ish

I had to make my own. There are a couple tutorials online if you Google “make glove box” or something to that effect. Generally it’s holes cut in a box with gloves

I wonder if a simple method for submerging equipment and smallish flasks for a sterilising soak within the glovebox, instead of trying to manipulate slippery items in the cramped space? An immersed saturation soak sterilisation method might well correlate to lowered infection rates. Buoyancy could be managed with a weighted perforated top screen to hold down? Or perhaps an inverted container containing the smaller flasks and sterilising fluid resting in a shallow dish? Just casual thoughts and ideas. Thx again Dustin!

About the oxygen peroxide, I have my metod : I make 95% + 5% bleach, it work perfectly, if you use it for 3 to 4 minutes maximum. Unless it would seriousely deteriorate the seeds, and i wash them 2-3 times with sterilised water. And i do the process inside the glovebox.
Be careful mixing peroxide and bleach. Once you start mixing chemicals like that, you run the potential of creating some toxic fumes.
