Have you tried magnesium, zinc, B vitamins, lots of red meat, and nuts? I also eat handfuls of dark chocolate (NOT milk chocolate or candy) on a daily basis. They might help.
@Edward Grand I definitely eat enough chocolate! I'm over 40, so I take my vitamins. I also found yoga and meditation to be a big help. Also, micro-magic-tea about once a week.

Really appreciate your lab time videos. Your stream of consciousness dialog exposing your thinking is both really entertaining and invaluable. I know you are concerned about talking during certain procedures and this might sound like a dumb question, but was wondering if you ever did a voice over? Since you don't have any head shots in videos like these, it makes the process dead easy. I know some of the impromptu charm might be lost, but you also cut some of your own risk. It always kills me thinking, he might be ruining his own projects just to get us more vid. Thanks a ton!
I've never really liked the idea of voice over. Too much work. I'm not into editing. Maybe later, but likely someone else will fill that niche.

Been using the Grand Tour Method to thin out the spores from a tuft of cotton. It REALLY works.
The zig-zag path thru the agar (agar-auger) generates so many separate colonies! I have a scope now, so I can verify if these are

I used the technique for like 5 swabs the other day...gotta go try to pick some dikaryons. I think they're past the mono stage already. It's amazing how one swab can generate so much work! Fun work though :)

I can't put into words how thankful I am that you are doing a series of videos like this! This whole process can be very hard to explain to people. The term pseudo-rhizomorphic still confuses me hahah
It still confuses me too... I had two monos yesterday that I was convinced weren't monos. I checked each of them for the third time for about 20 minutes. Not a clamp in sight! If I didn't have a scope, I would have sent them to grain. Made that mistake multiple times now!
@@edwardgrandThe wording itself is what confuses me. Pseudo-rhizomorphic is a cultigen that appears rhizomorphic but is not?? Im not seeing how the pseudo prefix is describing anything accurately since the term rhizomorphic describes the morphology of a colony and not the number of nuclei it has. Would mono-rhizomorphic be a better term? I feel like there might be a better way to describe this phenomenon….
@@cubertbensin Blame it on 'Judge Doom'. He is the first one I ever heard use that term ;)
The only problem I see with mono-rhizomorphic is that it implies there should be a 'di-rhizomorphic'...and even a mono-tomentose and di-tomentose if we are gonna be inclusive. Remember these are generic Mycology terms that are not exclusive to our babies. There are many species that could be described a 'di-tomentose'. I think it might just get more confusing to introduce new terms. Maybe we should bring this up on the Discord? Would love to hear other's input.
@@edwardgrand We should definitely bring people in on this and see what they think, I wonder if others are having a difficult time with this term or if its just me. I think the term pseudo-rhizomorphic carries that same implication we are worried about. If we are calling a monokaryotic cultigen that expresses rhizomorphic traits ‘pseudo-rhizomorphic’, wouldnt that imply that a ‘genuine’ rhizomorphic cultigen is dikaryotic? (I use the term genuine here because pseudo means ‘not genuine’) I agree with your point when we had our previous discussion about this, I think we can drop the prefix and just call it rhizomorphic.
@Cubert Bensin I wonder if there are other species that have monokaryons that can exhibit rhizomorphic growth? Armillaria is known for it's rhizomorphs. Don't know about it's monos. Seems like I saw a paper years ago? Maybe they have some proper terminology?

With my ghetto cross I have been taking pins from all my plates aswell especially crossing verified monos if you start seeing pins inside of the plates they definitely have formed dikaryon
Don’t sell yourself short. VERIFIED Monos are higher-rent subdivisions, not “ghetto” 😂
Can you do a video talking about pressure cookers and autoclaves. I heard in one of your videos that your cooker goes up to 12 PSI and am super curious which one you use. Your channel needs more subscribers, the way you have motivated me to not drop the hobby is insane. Thank you again and please keep making videos.
I'm in Asia, so I don't have access to the 'normal' brands of PCs and autoclaves. My main goal was to motivate people not to give up. TBH, I go through the "F..k this, I'm gonna quit" cycle about once a month. I'm probably on my 5th round of 'restarting' in the last 30 years. I think everyone does. It's not all peaches and cream, but we have to motivate each other to just take out the garbage and fire-up that next PC cycle!

You're plates (3,2,1) at the top are blocking proper flow. That's why it's creating turbulence in the laminar flow

How would growing them on a slide be of benefit? Petri dishes can be viewed under a compound scope

@@arctictimberwolf Clearly you have a lot of work to do on your anger issues before you worry about viewing mycelium. I hope you aren't abusive to those around you. It seems like you have a tendency to lash out unnecessarily. I'll try and answer your odd and nebulous questions. The thickness of a petri dish is difficult to answer due to the ambiguity of your question. Are you talking about the entire dish or the thickness of the glass or plastic used to construct it? Both dimensions would vary considerably depending on material and manufacturer so if you clarify I can then provide an answer. The neat thing is that almost all petri dishes are made out of clear material that you can actually see through! Next, YES! I DO own a few microscopes! I have an Olympus and a Swift compound and stereo microscope, respectively. I haven't found ANY issues in viewing mycelium or spores under the microscope in or out of a petri dish so I am baffled as to why YOU do. Perhaps you need to take a course in microscopy? In answer to your third question, yes! I CAN calculate focal length. Your fourth question is again poorly worded and not very specific but again, yes! I CAN understand the "meaning" of the numbers (N should not be capitalized) an "eye doctor" writes on a prescription regardless of whether they are an optometrist or ophthalmologist. I guess you were just taking a survey to see what skills other people have that you are clearly lacking. Were you looking for a tutor or a mentor to walk you through the process? Fortunately there are many videos out there on youtube that can probably help you learn to use a microscope and even how to make your own slides!
Best of luck in your microscopy and anger management courses and remember, don't let anger and violence get the better of you. Take a few deep breaths and meditate before you harm yourself or others.

Could you please make a note to briefly explain what happens with a Di/Mon cross? I asked in a previous video, what becomes of, say, a lone manokaryon among two dikaryon cultures in a spore plate?
You should look up "Buller Phenomenon". It is well-documented, but if I discuss it here, it will confuse people. You can find me on SM if you want to discuss it personally.
Is it really this simple to do crosses? Im sure its more nuanced than im understanding but i never really tried crosses since people make them seem really difficult using things like snake venom.

No. It's not necessary. We are just trying to dilute spores, not count them. If you do a 1X, 10X, 100X, etc., you will have one or two plates that are at the right concentration. If it's too many, use it for a dikaryon or backup. Too little, reuse it for the transfers of monos. ;)
Can a Monokaryon fruit?
Nah needs to pair and form clamps to become a dikaryon

Seems like it would be easier and more productive to just choose dikaryote mycelium from the plates, grow it out, then clone the best of the best fruit bodies instead of dealing with monos.
It is if you just want fruit. This kind of thing is for those who like to play/control a bit more and try to create unique combinations.
Perhaps a surgical mask, for when you're speaking to us? Just to reduce airflow.
