    Hello Sir. I have some queries about plant collection and drying processes.
    1. Can we pluck leaves and leave the plucked leaves to dried separately?
    2. Can we cut the roots and leaves separately and dried and powdered at the same time.
    3. How long time we should dry the plant sample in shaded area?
    4. After powder prepared, how long time we can use powder to carry various experiments?

    Thank you for the clear explanation. I have a question- after methanol extraction and evaporating the methanol, when you re-dissolve the plant residue, can you use any solvent other than methanol? Can I re-dissolve the residue in DMSO to make 1 mg/mL extract in DMSO? Will it dissolve in DMSO? For my experiment I cannot use methanol. It must be DMSO that's why I am asking. Thanks.

      It depends on the work, is it Ph.D or Msc project. Ideally we take 2 polar solvents and 2 non polar solvents. You can increase if you have time and interest.

    Sir I have a doubt . I was asked to take the quantity of the sample to be dissolved in the solvent according to the concentration of the sample. Can you tell me how to check whether the sample is concentrated ? Is it by keeping the sample in the solvent i.e; 1:10 ratio overnight and if the solvent evaporates to 3/4 th it means my sample is concentrated Sir?

      Ziplock will do or any air tight vials and refrigerator (freezer).

    Sir how can we calculate the amount of herb needed for a certain amount of liquid extract? E.g. if we need 0.5 litre of extract for storage then how much herb will be needed for extraction?

      For 1 ml of extract usually 20-25gms of dry weight is required. Its all trial and error cannot be predicted.

      As long as you can regulate the temperature any oven is fine. Keep for 2-3 days at 40 degrees

After extracting with all solvent...What will v do..we will calculate % yield for all extract or what?       Consider the oil directly to be ur sample and proceed. But i don't know if this will work i haven't worked on oils. Try reading papers on oil or u can ask some experienced person in research gate.

      You have to dissolve it in known volume of methanol so that you know the concentration. If ur experiments require crude extract in solid form then u can leave as it is and store it
