Replace the amount of tinned pumpkin with an equal amount of home-cooked squash. Kitchen scales would be the only way I know of getting an accurate result.
It's actually hard to say because store bought pumpkin is typically the same in moisture content-whereas if you make it at home they seem to come out a lot more wet

π sugar dissolves with the liquids, whereas the flours and such will need to be slowly incorporated to avoid dry spots in your batter
