    At 15:48, you wrote that it is positive supercoiling, how can it be +ve, if the Lk is decreased from 9 to 6, according to your discussion it should be -ve supercoiling. I didn't get it.

    I don't understand unfortunately around 12:23 you said DNA has a right-handed (clockwise) helix having a right-handed supercoil (clockwise). That is right-handed helix structured DNA + right turn causes positive writhe right?
    And you said around 19:33 "positive supercoiling = left-handed turn = clockwise direction". But why left-handed? shouldn't it be right-handed as you said before?

    Sir here your tell us that linking difference = l- l0 but actually it is l0- l is the linking difference i think so... So the. Linking difference us +3 not -3 so it is positive supercoiling 🦹🤔

    Another question - When a supercoil is formed as a result of double helix unwinding, topoisomerases fix that and remove the supercoil but the unwounded helix does not necessarily return to its initial helical state. Thus we get diffence in linking number.
    This is how i get an image of DNA supercoiling after watching the video. Is it correct?

    As the writhing number (Wr) can be both pos and neg and both increase or decrease the twist number (Tw) as the linking number (Lk) does not change in ccc it's all a matter of the Lk when Wr is 0 which equals Tw and if it's overwound or underwound with respect to Lk naught which is the linking number of fully relaxed DNA under physiological conditions which is usually bp/10.5 and this is what is changed by the Topoisomerases where the Topoisomerase I changes one twist and Topoisomerase II changes two twists (by changing writhes), right? So if delta Lk (Lk minus Lk naught) is negative the DNA structure is negatively supercoiled and has fewer twists when Wr is 0 and if delta Lk is positively supercoiled it has more twists when Wr is 0, right?

    Sir I didn't get it full if we talk about counterclockwise writhing,why doesn't the number of twists decrease?
    Dna here has right handed twisting originally and upon being negatively super coiled why does its twists increase, shouldn't they decrease as it'll open the twisting??
