Is there a point in beating them on normal mode ? Or should I just go straight for hardmode
I'd say go for hard mode, there are better unlocks there
Why are some completion marks bloody? Does this mean that I have to beat them on Hard? If you beat them with Hard mode on do you unlock any more items?
Yes, take Bethany as an example. In order to unlock her, you must win with Lazarus without losing a life on HARD mode. Lots more items are locked behind beating a boss in hard mode.

amazing video! but my hard mode completion marks aren’t showing up and im not sure why. it doesn’t show up for hush, delirium, or mega satan. it it because you have to beat them in normal mode first? pls let me know if you are reading this.

is beating mother only available through repentance DLC? I'm on mac it doesn't support repentance on steam
One thing is if you get the hard mark first you don't have to do it on normal.

I don't understand how pro Issac players casually fight Mega Satan as The Lost. I barely have any of the Hard Mode marks but have most of the normal marks for the characters. Now it's just unlocking Keeper for me and killing all bosses as everyone on hard. (Excluding Mom's heart/It Lives for Lazarus).

Why is it that I beat Mother on hard mode, but I did not get the mark, or achemvents. Just the ending? I tried it again with a different random run and still did not get it?

Lilith is super strong in greed mode because she can stack box of friends a bunch. However, she can't do this as easily in the ultra greed fight itself. My strat is to coast through the eariler levels and prioritize charge items (car battery, the battery, habit, etc.). You need to win greed as Azazel though.
Cain gives great early starts because he starts with a key, allowing you to reset 2 pedestals at a time for a broken start.
Tainted cain does this easier, because steam sale is cheap to make. With 2 or 3, you get more money than you spend on items, free win!
Azazel is an all-round good character who makes the early game easy for long enough to get a broken run going. Plus, he unlocks Lilith.
The Lost isn't the worst. He's still, yknow, the lost, but he can afford curse rooms and fly to get any pickups in the greed shop, which can snowball decently.
Isaac starts with a bomb, allowing you to bomb keepers in the shop for nickels, steam sales, or head of keeper.
Judas can afford a discount key with his starting 3 coins, and, in Repentance, Book of Belial stacks, allowing you to snowball like Lilith. Again, get other ways of charging it! 4.5 volt is a free win

On Rebirth you can't kill Hush, Delirium, Mother, Beast and Ultra Greed
