When Dean gets a glimpse of Sam’s heaven memories. How devastated he was to see Sam’s greatest memories were being independent. And him throwing away the amulet at the end.
I have to say the level of devastation and "I give up completely" is strongest in the scenes when he loses Cas in s12 and s15. Like. Usually he's angry and pumped on adrenaline and wants to immediately fix it. But when he loses Cas its like the pain and heartbreak completely paralyse him. I felt those moments so hard and Jensens acting is always so on point.
I think the scene with Mary should be included in this list. When he tells her he loves her and hates her, how he had to be a brother and a father to Sam. That scene is so full circle and heart wrenching because it pretty much sums up all of his feelings and the fact that Mary's decisions lead to what happened to them. Or when Dean finds out his dad died because he sacrificed himself in order to save him.
When they lose Charlie to the Steines... As much as he blamed them and Sam, it's written all over his face that he blamed himself possibly the most, as she was doing research to help him.... Jensen Ackles, you brilliant sexy tear-jerking bastard 😢😢😢 I don't cry, ever, at anything else in 20 of my 33 years of my life, and somehow every single time I watch him and Sam during the funeral scene, suddenly, years of stone facing my way through life give way to a frickin waterfall of emotion!!!

If you're not crying I'm crying, when Dean was ripped from Hellhounds I cried for 2 years and stopped watching Supernatural until I got up the courage to watch the series again! 😭 Give this man a break, he's been through a lot during the series! Give him a hug, a pie, beer, a warm blanket, and a teddy bear! 🤗🥧🍺🧸
He had to be the parents and big brother. He’s the only fictional character I can think of that has been through the wringer. John is responsible for it too

Cass speech in season 15 is my 2nd favorite of that season. Sam's speech to dean about only knowing one thing that his brother will always take care of him was the best.

To me 5 doesn’t count because of all the contradictions that goof the storyline in season 15. That includes the empty taking Castiel.
