    There's away to, when you're recording your videos to make them turn out really loud after recording. You either turn your volume down low or turn it up high to make your videos have much greater sound playback! A viewer can always turn the volume down if it's to loud, but can only turn it up so high when they simply cannot hear it, like this video here! You put in all that time and work making a video that the viewers can't even hear, that's frustrating mostly for the viewers! I hope you understand why I wrote this.
    It's just to bring to your attention that I do like your work, but I like so many millions of others can't hear that well and are trying to figure out how we can in some small way help. Hopefully you're next video will be so loud, I will actually have to turn it down, lol. Thanks!

    when you squirt the LC on the agar do you roll it around or just let it sit in a droplet?

      Great question! Rolling it around a bit is a good idea, but either way it'll do the trick!

      Good work! Some buildings are more problematic than others so it all depends on your unique conditions. We had a lot of $ on the line and contamination was not an option so we take precautions! Mush love!

    Can i use a very small grain spawn instead of the agar, like 2 grain thick??

      I suppose so, if you can make it work, but agar will give you a more accurate panel of the organisms. For example, trich would be easy to identify on grain, but bacterial contamination would not be. Agar gives you a much clearer idea.

    How do you destroy the trichoderma petri dishes, isn't that special waste that has to be disposed of in a special way so that the environment is not polluted? Or is it simply burned in the waste facilities ? Thank you :)

      Nope - trich is everywhere and actually a wonderfully beneficial soil fungi! Throwing in the compost is good :

    After you squirt 1 cc of your liquid culture on to your agar plate, do you put it in an incubator? If so, what temp and for how long before you get some reliable growth that you can differentiate?

      We just store it with the other liquid culture jars, so in a cool dark room, about 67-69 degrees. You can see growth as soon as the next day, but 3-5 days will express a true panel of the lc.

    So are the ones that are not infected but are "not as happy" just no good and need pitched or just need more time? What's going on with those?

      They are definitely still viable and up to you if you want to use them. The slower growing ones can also be because you deposited less mycelium on the agar, just try and be consistent and observant to make that call! if they are growing a lot less slow than the rest, we tended to just dump them. Usually however, as long as they were not contaminated, we used them and they worked great.

      Yes, you could if you have to! One tip is to keep the contam BEHIND the clean portion in relation to the flowhood

    So, I assume the liquid culture contains some kind of grain-based sugar extract along with spores that have germinated that have colonized into mycelium and the agar in the dish also has some kind of malt extract nutrient?
    Just want to be sure because I plan on moving towards LC and I don’t want too much sugar with toxicity being an issue of course.

      The LC recipe here is just RO water and honey! We did start adding a teaspoon of malt to quicken growth, however.
      We also did not work with spore, but rather a single colony transfer from agar to liquid. If you are using cordyceps, a table spoon of honey per quart is a great recipe. Cordyceps are sort of sugar addicts and seemed to thrive the more nutrients we added. That said, increase little by little and monitor haha

    Hi, do you need the flow hood and have to wear all of the sterile gear when doing the testing? Or was that for when cultivating the liquid culture? Also, is there a need to test liquid culture you bought online from a reputable source? Thank you, love what you do!!

      a flow hood is very helpful but you can get away without one if your space is clean and you are still. The gear is not necessary for this test. If bought from a reputable source I would not worry too much, but still might be smart to squirt some on agar as back up and just to be safe!

      Yes you can totally use the lc tests as a normal petri plate and expand as usual. Just a warning with cordyceps, they will senesce quicker with transfers, but most other basidiomycetes should be just fine.       We have no used Karo, but I see many recipes with Karo so I believe it will work well!

      We bought a premix, mostly to save time. Fungi Perfecti has good medium, as does seaweed solutions! If you want to make your own from scratch I suggest skimming TH-cam a bit more, theres lots on here.

    you advise to use just plain malt extract agar to test your liquid cultures yet on the petri's you used to show in the video it says MEA (the antibiotic agar)
    so thats a funny little mistake there

      Nope, MEA = malt extract agar
      we mark antibiotic as MEA+

      Nourysh nope, a sister organic farm does that for us. It’s pretty easy to do. They grow in supplemented sawdust bags and if you cut slits in the sides they will grow out.

      Yes, very possibly there was not enough mycelium. We have just noticed that this is also an indicator of less robost cultures. :)

      To sterilise you can preheat the oven on 300°F and put your jars in there once its sone pre-heating, and once it starts bubbling its done, it takes about 50 minutes

      The vectors are many. Dirty cultures, extremophile surviving the cook, imperfect technique.... its rare for us, but happens occasionally with wild clones.

    My mate had a setup like this. I put a sandwich inside his laminar flow box and he never found it but quit 6 months later because everything he had got horribly contaminated and he could no longer fulfill orders.
