Hi miss! at 47:00, are we meant to insert the enzyme gene into the plasmid? isn't it meant to be that the plasmid would already have the gene that codes for lactase and we're adding our DNA fragment to it to see if it disrupts the lactase? It does say that on the image you inserted but the text says otherwise... just checking with you if you see this!

At 46:40 is it colourless to blue or blue to colourless?
the text says from colourless to blue so thats why im confused

from colourless to blue, if the fragment is present then it stays colourless as it stops the lactase gene, however if the fragment is not present then it will turn blue as the lactase gene will work
Hello, thank you for this video! I have a question - at 36 minutes you talk about the modifications being made so RNA polymerase can work properly. Why is it RNA polymerase, not DNA? What is the RNA polymerase being used for? Thanks!
DNA polymerase is used for semi conservative replication ONLY. whereas promoter initiates TRANSCRIPTION hence only transcription factors and RNA polymerase can bind to promoter. If DNA polymerase binds to the promoter it will technically inhibit the binding site of the promoter

I apologise that my comment is not related to this video. But I wanted to ask in regards to the ELISA test, which is explained in the Monoclonal Antibodies video. After adding the second antibody are you supposed to wash again to remove any unbound antibodies because I believe that that was not included in the method of performing the ELISA test
Yea u wash after the second one otherwise when u add the substrate next it will break down regardless

To clarify: When do you use DNA polymerase or RNA polymerase? (how to remember the difference?)

DNA polymerase = for DNA replication
RNA polymerase = for transcription

Great video! For the marker genes, I though that if the plasmid was taken up by the host cell then they will express GFP or they will be resistant to the antibiotic as they contain the marker gene (instead of disrupt the gene so it isn't expressed) or is it the opposite way round?
Hey its the opposite way round, as explained in the video..However, read the exam questions carefully to see the information given on where the DNA fragment is inserted.
