How important is working in sterile conditions while preparing the PCR Mastermix etc.? Working on ice in sterile conditions could get a little tricky I suppose?
+Libe Jenkins first we add the larger volume, it can be buffer or water, depending on the reaction. Then we add the lowest volume, and the reason is because in case of lower volume such as the DNA polymerase, it's viscous, so there will be a lift over inside the tip after we add the desired volume into the reaction mixture, If we add the buffer and the water before the DNA polymerase, we can the desired volume of the DNA polymerase and then washing the left over inside the tip by mixing the tip up and down with the buffer and water in the reaction mixture.
