Very well represented. However DNA binding domain of Zinc finger nuclease binds to specific nucleotides and not gene. Also recognition is towards nucleotides and not amino acids. Wildtype Fok1 endonuclease is not completely non specific. There is specificity with respect to its DNA binding at the 5'-GGATG-3' recognition site but DNA cleavage is non specific, 13 nucleotide upstream and 9 nucleotide downstream to its binding site. The Fok1 nuclease domain is used here.

Yes. You are right. The video only explains the concept without going into the details. The words gene and amino acids instead of nucleotide just makes it general to understand the overview. Whenever we talk about specificity it is obvious that the specificity is towards the sequence meaning towards the nucleotide. Thank-you for your feedback. Really appreciate it. 😊👍
