Hello. I would like to ask. If I would want to extract mushrooms, is it better to use soxhlet extraction setup or sonicator? With soxhlet to make water extracts I would need to boil water to get water soluble compounds. With sonicator it is only possible to extract alcohol soluble compounds and I will need to make boiling after also to get water soluble compounds if I want double extraction? Or sonicator can not only make alcohol extraction but water extraction in cold water without boiling too?
@SoloSp33d When extracting mushrooms, the sonicator is the superior choice over the Soxhlet extraction setup for several reasons. Firstly, the sonicator operates at lower temperatures compared to the Soxhlet, minimizing the risk of thermal degradation of heat-sensitive compounds present in mushrooms. In Soxhlet extraction, the extracted compounds are exposed to heat for a long time in the heated flask. Secondly, the sonicator provides better control over the extraction process, allowing for precise adjustment of parameters such as amplitude, power, temperature, and duration to optimize extraction efficiency. Additionally, the sonicator has a higher capacity, enabling the processing of larger volumes of mushroom material in a shorter time frame.
Contrary to the misconception, the sonicator is not limited to alcohol extraction alone. It is capable of extracting both alcohol-soluble and water-soluble compounds from mushrooms, including polysaccharides and other bioactive components. Furthermore, ultrasonic extraction in cold water is possible without the need for boiling, making it a versatile and efficient method for extracting a wide range of compounds from mushrooms. Therefore, the sonicator is the preferred choice for extracting mushrooms due to its ability to operate at lower temperatures, offer better control over the extraction process, and accommodate higher processing capacities.

How is the amount of solvent determined? Thank you, I’m having problems with a batch I think I added too much water to the flask
@momsmushroomsjodyfoster5786 There are two aspects:
1) The solvent volume should be high enough so that the flask does not run dry when the solvent evaporates and fills the Soxhlet well. The solvent volume should be low enough to allow evaporation. Our suggestion: At the start, fill the Soxhlet well manually with solvent to just below the apex of the siphon and fill the flask to the level of its maximum diameter.
2) Depending on the compound amount to be extracted, you should not oversaturate the solvent in the flask, which would cause separation (e.g. precipitation). If you see oversaturation, use less botanical material in the Soxhlet well.

Thank you so much for your fast response! It’s difficult finding answers for my questions! But, again please. Should I be adding 250mills of liquid to the a 500mill flask? Because the liquid expands as its heated, correct? I tried adding more than 250 mils of water, and it was disastrous.
@momsmushroomsjodyfoster5786 Fill the flask to just above the widest diameter. Depending on the volume of your Soxhlet apparatus, you may have to add a little more, if you see the level in the flask falling too low.
Make sure that your condenser condenses all water vapor, so that you don't lose water during the process. You see that our Allihn condenser has a very narrow opening on the top to avoid vapors exiting instead of condensing. Use a low coolant temperature on the condenser.
The expansion of liquid due to heat is insignificant.
Use boiling stones to get smoother boiling in the flask and use medium heat to start slow. Once you have everything under control, you can increase heat in small steps to speed up the process.
Alternatively use cold water and sonication and avoid the Soxhlet all-together.
In a Soxhlet extraction, the extracted compounds are exposed to heat in the flask for a long time, causing heat degradation. With our sonicators, you can extract at cold temperatures, so that the compounds are not exposed to heat.

How long of a process is this, lets say I'm extracting mushroom compounds. How will I know when it has extracted the most valuable compounds and will this vary depending on starting materials. Thanks

As explained in the video, in Soxhlet the extracted compounds are exposed to heat in the flask for a long time. That is not good for heat sensitive compounds, because it will cause heat degradation. So you are destroying what you just extracted. With probe sonication on the other hand, you can
- break cell walls - Soxhlet does not, and
- extract soluble and in soluble compounds - Soxhlet does not extract insoluble compounds
Most importantly, ultrasonic probe extraction produces great yields and works at low temperatures, such as -20degC. This preserves heat-sensitive compounds.

Can water be used as a solvent?
@momsmushroomsjodyfoster5786 Water can be used as a solvent in a Soxhlet extractor, but it will extract water soluble compounds only. Without sonication, the water or solvent will not break cell walls. Therefore, extraction is very slow and incomplete.
