Overview: Dr. Hooper studies how the gut microbiota changes during illness or disease and how it influences our ability to fight infections. In part 2, Hooper explains how a healthy gut microbes induce a host protein called RegIIIγ which helps to protect the host from infection by pathogenic gram-positive bacteria.
    Detailed description:
In this lecture, Dr. Hooper introduces us to the fascinating world of human microbiota; the microorganisms that live within our bodies. Although we may think that most bacteria are harmful, Hooper provides ample evidence that symbiotic gut microbes are important to good human health. Her lab is interested in understanding how the microbiota changes during illness or disease and how it influences our ability to fight infections. Using germ-free mice, they were able to demonstrate that a healthy microbiota can shape development of the host immune system and provide protection against dangerous infections like salmonella.
In the second part of her talk, Hooper explains how the balance of organisms in the microbiota is maintained. By comparing DNA microarray data from normal mice and germ-free mice, Hooper’s lab was able to look for genes induced by the microbiota. They identified RegIIIγ, an important protein involved in the protection against pathogenic bacteria. They showed that RegIIIγ forms pore complexes in the membranes of gram-positive bacteria and kills them. In mice and humans, the intestinal epithelium is coated with a layer of mucus. Typically, there is a gap between gut bacteria, which are found in the outer part of the mucus layer, and the epithelial cells. Hooper’s lab showed that RegIIIγ helps to maintain this gap by preventing gram-positive bacteria from colonizing the intestinal epithelial surface. This, in turn, prevents infection of the host.
    Speaker Biography:
Although she always was interested in science, Lora Hooper’s love for biology started after taking an introductory class at Rhodes College in Memphis, TN where she was an undergraduate. Hooper continued her graduate education in the Molecular Cell Biology and Biochemistry Program at Washington University in St. Louis where she joined Dr. Jacques Baenziger's lab. For postdoctoral training, she stayed at Washington University, in the lab of Jeffrey Gordon, where she began her studies of the interaction between gut bacteria and host cells and discovered that bacteria have the capacity to modify carbohydrates important for cell signaling.
Currently, Hooper is a Professor at The University of Texas Southwestern Medical Center and a Howard Hughes Medical Institute Investigator. She has established one of the handful of mouse facilities that have the capacity to breed germ-free mice. Using these mice, her lab explores the symbiotic relationship between a host and its microbiota with the aim of providing insight into human health.
Hooper was a recipient of the Edith and Peter O’Donnell Awards in 2013 and in 2015 she was elected to the National Academy of Sciences.

(On Saturday of April 22, 2023). On the Matter of PhD Lora Hooper of University of Texas Southwestern (USSW) on the Topic of Microbiota in Mammalian Gastrointestinal Tract and the Supposed Beneficial Symbiotic Relationship Therein: 1) Lactobacillus Genera and others are also Modulatory of this ever-Constant Threat of Death (Sepsis in a Perpetually Immunosuppressed Victim is Real) or can be pathogenic also along the lines of Salmonellosis or EHEC (N157:H7) just because there is the Dyshomeostasis of Imbalance, as Virulence only becomes evident then; and the Bacteroidetes and Firmicutes (Monera Kingdom) along with Yeast/Fungus (Candida albicans), and Methanobrevibacter smithii (Archaea) are as Pervasive as Foreign Proteins which are only dwarfed by the Transposition/Translocation of Viruses 10^15 within the Gut Microvirome. 2) If this Environmental interaction was absent, then and only then would Mankind genuinely be What Will and Self-Determination Sense there might be makes of such Creature. Otherwise, yeah a man is not only the Genotype-Phenotype but also what the Microbiome does therein, a true disciple of Determinism. 3) Antibiotic Therapy is to be last Resort Therapeutics (Lifesaving Quality) rather Convenience store Candy as has become with the Proliferation of Immunodeficiency (It used to be something like USA and now it metamorphosed to "United States of AIDS"). In the Event these Breathing and Talking Biohazards are not self-Annihilated, so will it be Nature to do so. I mean who would think Feces would be up for grabs these days--after all it would be far better available than Proton Pump Inhibitors (PPIs) or Chemotherapeutics of Antibiotics. It is wonderful to be absolutely indispensable anyhow; 4) Rather, one must rejoice at the Ecological Perfection and the Inherent, insignificant Extent of Mankind (notwithstanding its Microprocessor and Radiocommunications). When one contemplates Microbe-Free World, this Entire Ordained Setup is Nihil and intentionally eschewed, for the Mesenteric System would simply have to be what Man makes of it. As of Today, the Reversal of Input/Output Flow of Neural Information within is from the so-called Mesenteric Nervous System first then to Central Nervous System (CNS) when in the Homo sapiens "sapiens' cultivar, should have it CNS to MNS if anything. It does look like that when 80% of the Totality of Serotoninergic Signal Transduction comes from Microbiota Origin. The Brain's Impermeability might be Strong and Exceptional (Blood Brain Barrier), but when one understands Pathogenesis of Disease, one can also deduce the Enzymes of Biosynthesis are also capable of Entering where and when they Please (Enzymes that Degrade the BBB are Moot/Mute point these days). Inflammatory Bowel Disease, Irritable Bowel Syndrome, and Peptic Ulcer Disease are simply a Reigning and Prevailing Monera-Microbiota Bio-thuggery. My Proteomics' Understanding currently places Neurodegeneration, Osteoarthritis, and Heart Disease well within the Environmental Predominance of Bacteria (Starting within the gut) and Viral Aetiology (100% Specificity notwithstanding Human Genome Project Determinations and World Medical Guild Dictates). 5) I would implore only one thing. If a man chooses to be of this Quality, that such be Respected. As of Now, the Chronically and Diathetic Immunosuppressed Genotype-Phenotype has the total monopoly control of Policy and Treasury therein inventing Pandemias (Coronavirus Type), Diseases (Anxiety and Depression) and hoarding Public and Vital Resources for their Exclusive Ends/Means and Wants. I find this unruly and completely abject Tyranny for which I have admonished endlessly Apocalyptic-Proportion Consequences. The Majority much less a Fanatical Minority should not dictate anything, let alone enjoy and bask in the Loot of incessant Pillaging and Plundering with aid and abetment of Pseudoscientific, Pseudo-jurisprudential Machination. This is a Science Exposition surely, but it is also a systemic and prevailing Trend within unbeareable limits a civil Forum of an Open Society with Democratic ideals and the Hygiene Concerns therein. PhD Hooper, es wurdet besser sein wenn man nicht haette gemacht Die Krieg zu Finden Friede.
