    Dude, you lab looks grimy af. Maybe clean up? Also, you're making several mistakes that make your stain pretty worthless. Yes, it will stain the cells, but no, it will not conform to any standard and in result you will not be able to judge a number of characteristics correctly. Please don't use this for Malaria, for example!
    Here are some suggestions for improvement:
    - Don't use tap water! Use a buffer solution (Soerensen or Weise) at pH 6.8 for hematology and 7.2 for malaria. The uncontrolled pH and buffer capacity of your water will make the colors totally variable depending on lcation and time point!
    - Don't filter the stain after dilution. If it needs filtering, filter before. Filtering it as diluted liquid retains dye in the filter and shifts the color balance.
    -Fix with 100% methanol. Adding 30% water will just make the result worse. Many authors even claim that daily methanol change is required due to moisture uptake.

      tap water is not supposed to be used ,you can use rain water to improvise because rain water is compared to pure water since it doesnot contain those minerals

    Sir can I use 50 ul of giemsa stain and 450 ul distilled water for 1:10 dilution? Pls reply

      How long can we reuse the absolute methanol, and how do we store it so we can use it for more longer time?
      And can we reuse giemsa 10% as well after first staining?
