    Hi Dr. Raki. I work with beneficial fungi of plants, generally considered safe for humans. I am interested in characterising the extracellular extracts with ethyl acetate. How would you suggest to operate to obtain the culture filtrate? Can I work on the bench Or do I have to do this in the safety laminar? To separate the broth, do I have to use the filter sterilisation with syringes (to remove the spores as well), or can I just filtrate with a Whatmann filter paper and think about getting rid just of the mycelia? THANK YOU

      Its always advisable to work in laminar though ur sample is safe you can avoid contaminants from outside if your working in LAF. Filtering the spores would give you more exact information about crystals since your going for characterization spores might be a hindrance

      Yes it will evaporate all.the solvent then you can add known amount and redissolve. It's must if u do. For lower work at UG and PG we don't do so properly.

    Sir.. I have prepared plant extract using methanol and ethanol solvent.. Now I got semi liquid thick viscous extract... So, what diluent should I used to dilute this crude extract since it is not soluble in water.. Some said DMSO is best diluent but it is toxic.. Sir please suggest me what diluent should I used?       You should use the same solvent , methanol and ethanol only. Since u used these solvents only in the first

    My doubt is the I use the methanolic solvent to the extraction of plant material but after extraction how to separate the ethanol and my plant extract when I used the your technique that is not sufficient because in the extract ethanol also be present so how I separate

      Yes you can centrifuge them at around 10,000 rpm for 10 min and separate the residues and collect the oil

    Bro I made extract from seeds and it is an oil extract now how can I make its dilute solutions as in ur previous videos I have seen that if we have 1mg extract then we add 1ml solvent then what about oil?

      You can add organic solvents which are soluble to your oils like chloroform acetone. Check the solubility practically or by reading papers

      1:10 ratio for extraction. After evaporation add accordingly to make a known concentration. Watch the video on methanolic extract of plants for more info

      You can use hot air oven and dry it but the solvent will be evaporated so it can't be re-used
