His deadpan delivery is what really makes this hilarious.
Aww yeah, I was at this show. And that moment where she stopped and smiled at the beginning, she was looking at me :'D I was in the front row and kind of staring at her in awe and she saw me and I guess she found it amusing. I don't blame her, I'm a crazy fangirl and being around her turns me into an idiot.

I like her voice.
And I like the predominantly low-key performance.
So many singers want to make a "big production" out of it.
But these two have radically altered the melody - not for the better.
Perhaps the banjo (or whatever it is) can't do the "diminished" chords of the original.
And they have slightly altered Gus Kahn's brilliant words.
The audience reacts as if it had never previously heard the song.
A good, if not perfect rendition.
Knowing who they are is part of the context that makes it funny. She's a big star (mostly rock/cabaret) who often throws together ukulele arrangements of songs to do as one off performances, or even writes something quickly, for fun. He's a well known writer, a novelist not a singer. They'd only recently married when they did this tour of "an evening with" where they did really low key, intimate performances and readings of their works. Knowing the people is what makes the performance hilarious (e.g. the very stiff performance and unrehearsed "well..." comment from Neil) to those watching.
