    Great talk, dude. How much time did it take you to draw that board content? I had a few professors like you in college (engineering) and it was always such a delight to find one, succinct, accurate, and an overview followed by a bit, and then some serious "delving in" to the subject. Nice. ;-)
    I had one professor, for dynamics, that said just after he gave his name, F=ma, we're done...then proceeded to blow our minds with some serious calc and diff equations for things like Coriolis effect, etc. We weren't even REMOTELY done, it turned out, but yes, F=ma was pretty much the cornerstone of all of it.
    One question (from an engineer, not a biologist). When initial pluripotent stem cells, which by definition, can apparently become any body part are "differentiated" into say bronchial cells, are they then only capable of being "switched" to make only that tissue and any tissue that might follow (lung tissues maybe), or can they still be "switched" to make say, bone in the normal progression of the fetus? I guess what I am asking is, from what I can gather scientists "make" pluripotent stem cells from differentiated stem cells, but do (or can, ever observed) the stem cells in a fetus ever do this, revert back to pluripotent cells?

    • @butterbeer4582
      @butterbeer4582 4 ปีที่แล้ว

      Hello, from my (very limited) knowledge of first year med, it seems that what you are talking about is when a pluripotent stem cells becomes multipotent stem cells, which are "comitted to give rise to cells that have specific function, for example blood stem cells". The only case of cells reverting back I've heard of is alpha-cells (that produce glucagon) being converted to beta-cells (that produce insulin) in diabetic patients as a response to the destruction of beta-cells. Only 2 % of alpha-cells do this though, so it's not super effective.
      According to my embryology lectures, "the maintenance of cellular identity is an active process mediated by repressive signals that stop the natural tendency of cells to change";
      So the presence of insulin itself keeps alpha-cells from reverting, but when it's not present (in the case of diabetes), some of them are liberated from this restriction and have the ability to change back.
      Therefore in specific conditions, non-beta cells of the pancreas can be reprogrammed, and this is called cellular plasticity.
      Hope this helped, anyone feel free to correct me, and sorry if some of the words are inaccurate (I'm translating all of this info from French)

    • @MrJdsenior
      @MrJdsenior 4 ปีที่แล้ว

      @@butterbeer4582 Yes, VERY interesting. On the glucagon/insulin alpha cell reversion thing, I wonder if researchers are trying to figure out how to increase that percentage, CRISPR mode?
      My father is diabetic, as were several of his nine brothers and sisters , so the glucagon/insulin thing is really interesting. He is a physicist/engineer and his doctors told him NOT to try to self real-time prescribe his own insulin doses (which probably in almost all cases is absolutely the right advice, but you know what they say "idiot proof and engineer resistant"). He didn't heed their advice (and I'm AT ALL not advocating anyone else try this here, outside the given medical systems currently available) but he was able to work out a pretty simple equation based on meal calories (est., he got really good at it though), present sugar level, and a few other parameters (don't remember them all), and was able to control his sugar to a much greater degree than just using a standard dose (he did a LOT of research first, too).
      He recently went in for an ankle replacement at age 86, because he lost the ability to exercise because of it (walking, anyway), and he has been diabetic since age 40. He asked the doctor if he was in the 6 sigma age group for this operation by this doctor, and he said no, he had one at 88. Dad asked was he a 45 year diabetic? Doc said NO...good point. Because of his accurate sugar control over all that time he has restricted problems to a bit of peripheral neuropathy, a quad byipass 6-7 years ago, and a likely unrelated hernia about 12 years ago. He takes no medication, just insulin.
      Interesting aside, one of his best friends went of to work at a bio-engineering co. and they made the first tested COMPLETE artificial pancreas, fully self contained except a port to refill the reservoir, and a subcutaneous battery box for relatively simple replacement, when they ran out of charging cycles.
      Apparently, they worked spectacularly, and one of them is still in one recipient some 20 years or so later, working like the day it was implanted (these were prototype units not designed to operate for nearly that long, some relation to a couple of the Mars probes and the Voyager crafts. I'm not sure why it wasn't brought to fruition, but I worked a tracking laser cornea shaper for SPECTACULAR vision correction that didn't require the in my opinion, not smart flap cutting done for LASIC. I know when we were about finished with the FDA approval hoop jumping, we were sold to one of the major manufacturers (who made fun of our product all the way up to the point when it very much looked like approval was immanent), and who then put it on hold for a long while, because they didn't want to re-tool to produce it. The doctors didn't like it because a doctor wasn't really necessary for the procedure, other than as a fail safe observer (actually not at all, litigation problems aside) and it was going to be a VERY quick, very inexpensive procedure, with VERY minimal very short term discomfort, better than LASIC, and with better visual outcome, as the cornea was quite measurably a better surface, all around, than pre-"op". We were also, at that time, talking about a laser retroreflective "power" mapping of the system to correct all aspects of lensing and fully compensate retinal shape, as well. Thx for the info, good luck with your career, and cheers. ;-)

    • @butterbeer4582
      @butterbeer4582 4 ปีที่แล้ว

      @@MrJdsenior Hi, I don't think gene editing for that mechanism would be that useful since type 2 starts with insuline resistance, which is mostly due to fat getting in and "blocking" the insulin receptors. The beta cells then overcompensate by producing more insulin for a while, but eventually get irreversibly exhausted.
      So it's probably more useful to focus on the resistant cells, although there is a concept of trying to make the remaining beta cells more effective and function longer. This is the limit of my knowledge, you'd probably have to research this more :)

    • @butterbeer4582
      @butterbeer4582 4 ปีที่แล้ว

      Thanks, that's all very interesting to me, I'm actually studying to be a biomedical scientist not a doctor, so aspiring to work in the same kind of field!

    Hi, really enjoyed the videa and that it broke everything down into easy to understand steps.
    Can I just clarify the timing? You say 4 weeks but then 16 days for the respiratory diverticulum forming; 4 weeks is actually 28 days. Which is it?

    • @minass
      @minass  5 ปีที่แล้ว

      Matthew Cruice it’s 4 weeks. Thanks for spotting that one. Where is the time stamp so I can add a correction?

      @@minass It's at 3:00 . Thanks for clarifying.
