  • Why did an adolescent boy start growing breasts? And how did PCR help scientists solve the mystery? This video explains what PCR is and how it works - all in the context of how a team of scientists diagnosed an adolescent boy with an XX + SRY genotype.
The research story from this video is based on this paper:
    M. S. Kim, P. H. Hwang, D.-Y. Lee, A 46,XX Male Adolescent Presenting with a Chief Complaint of Gynecomastia. Pediatrics & Neonatology 56, 357-359 (2015). doi.org/10.101...
In addition to using PCR, the scientists used Gel Electrophoresis to determine the patient's genotype - here is a video about Gel Electrophoresis:

Thank you for your video!! Your example helps me understand so much better in detail!
I have a question on SRY gene replication using PCR. How will DNA polymerase know when to stop and only replicate SRY+ gene?

Good question! The answer has to do with primers, but it's hard to explain. I suggest watching this excellent animation of PCR to see why only the DNA of interest gets amplified th-cam.com/video/JRAA4C2OPwg/w-d-xo.html

What would the optimal temperatures usually be for DNA denaturation, primers binding and DNA polymerase ?

How do they know where to set the two primers in order to get the copys of sry? If they knew were sry was already why did they have to test for its presence?

Great practical info. Easy to understand. However, the final gender comment can confuse vulnerable minds. Keep up the good science without the social opinion.
