First episode of season 1, Gary's fanclub took him away in a car.
Cars have been popping up occasionally since gen 1. Mostly in the background, sometimes driven by police officers or side characters. Team Rocket has sometimes been making their getaways in non-balloon vehicles since season 1, including things like vans. Ambulances have been shown. As the other comments mentioned, Gary and his fan club have been travelling by car since the very first episode. And one of the movies did involve an RV.
The episode in season 1 where they find the kid who thinks he's a kangaskhan, I'm pretty sure the officer they meet has a jeep. And one of the gen 4 movies has a character who drives a car.
This may be the first time the *main* characters have really been shown to use a car, but this isn't the first season to feature cars in general.
