It's subtle, but a lot of the difference comes from the low end. 2:53 and 2:58 are probably the best examples of what I'm hearing from it. Hope that helps!
Sounds about right. You don't need a TON of gain to get a good distorted tone. More gain on the amp can often make the tone more "muddy" and "inarticulate". Boosting the signal before it gets to the amp, e.g with an overdrive pedal, which he has also gone over in one of his videos, or by playing with more attack/force, gives the amp more to work with. So by running the bare minimum of gain on the amp itself, you will often find that notes can stand out more by themselves.
This plus a lot of these amps have a silly amount of gain on tap, you get to a point and you're just trading dynamics for noise on top of your signal. Often an overdrive before the amplifier gives the amp a better signal/information to work with, by reducing the guitars low and highs slightly while compressing the sound and bringing up the mids a little can improve the articulation and tightness, plus help you fit in a mix better.
