Cool video. Btw Lasius flavus are not in North America so those are probably Lasius brevicornis or nearcticus(probably the former). Also, eggs will die during hibernation. Usually, only small larvae(1st instar) will survive.
You could split a wooden skewer in half so the end is frayed and then put the frayed part on the Cotten and twist it to get a hold of it and then just pull it out

Congrats on 1000 subs! To answer your question about the cotton that's stuck in the AC test tube... I always use long tweezers to remove my stuck cotton balls. Takes a bit of a steady hand as well.
I have a camponotus pennsylvanicus colony, how far into hibernation should they be in right now? (I hibernated them a week and a half ago) Also amazing job on 1k!!!
i have an 8" long tweezers in case the cotton is too deep. I submerge the whole test tube in soapy water (i put something heavy on top so it doesn't float), I try to pick at least one little piece of the cotton using the tweezers, and I start to rotate it as much as I can, this will relieve the pressure of the cotton against the walls of the tube with time, then I pull; sometimes you will get a chunk of the cotton, other times the complete cottom will completely get out or in the worst case you don't get anything out and you have to repeat, it normally takes between 5-10 minutes, if I don't see progress after 10 minutes I submerge the test tube again while I try with another one.
Go in between the test tube walls and cotton ball. With a sturdy skinny tool like a screwdriver or chopstick. And pull it out once you have a good hold f the cotton ball with the long skinny instrument
You should probably only touch a corner of the nest with a heating mat/cable instead of just putting the whole nest on the pad.That way they have a gradient

It really depends where you live. Once you see the ants outside hibernate then you know it’s time.

Thats what i do and it works for alot of ants so it they need cold or warm they move the brood
To remove the cotton, I use very small and long scissors, and pull it out by latching on to it
Do phidole queens need the same sent like meat ants or can they have any queen
