if itsn't visible moisture on the ground temperature is 15c but at 32000ft its -32 or higher and its clear and its night, we need to activate? if its clear day and sunny but -25 degree we willnot engage but if it is night do we need to active if there are no cloud or moisture

During the flight if there is no visible moisture or clear sky then no need to activate anti-ice. Day and night same rules, we can identify at night either with weather radar or landing light to identify the moisture.
