Coagulase test is done for identification of Staphylococci. Both slide and tube coagulase test has been explained in detail.

Why we have to incubated normal colony before adding the specimen ( pooled human plasma) ?

No need to incubate in slide test. But you have to incubate in Tube coagulase test for the enzyme production and action on fibrin.
