You can do this much easier. Fix the pipe to your bench. Put a pipe with the same OD aside, then scratch a line by holding a ruler across the pipes and move it along the pipes. Ready! Quick and easy and no measurement errors.
Clamp pipe or bar stock in vise rub a bit of chalk on the top of the item. Place level across the top of the item drag the level through the chalk in a level position and the clean line that you made through the chalk is top dead center, takes but 30 seconds.

I would probably put the square on top of the pipe letting the "handle" part touch the side of the pipe and using the spirit level to get it horizontal. Then just use the scale on the square to mark the spot.
if the "handle" part is long enough (could be short for larger od pipes ), can also put spirit level on the side of the pipe and measure center of the pipe
Put a mark on the pipe using the square when level put the square on the other side of the pipe and do the same thing marking the same spot and the just split the difference. It take less than a minute.
If its the first branch the center is anywhere you put it... think about it... no matter where you put a hole, that can be the center. There is no off center on your first branch.. too much left or right, just turn the pipe in that respective direction and boom! Center.

I have no training in plumbing whatsoever but I think it would be easier to simply trace the radius of the pipe on a paper, then fold the paper half horizontally & half vertically then use the paper as a template making sure you clamp the pipe firmly on the floor so it does not move. With the template, you can easilly find any degre angle of the pipe. After all why complicate the process?
Get rid of the square just use the level against the pipe in the vertical position measure half the diameter from the level to the top of 4" then mark or use a center finder
Just use a paint pen draw a line perpendicular with the pipe where you would like it the pipe to intersect then use a level and put it across the pipe perpendicular slide it across the pipe and now you have an intersecting point that is dead center

Unless there was a seam and let's say the seam was supposed to be at 136 degrees so you find your seam location and measure back to zero then center it.
Too complicated for a simple task. Just measure the OD of the pipe, wrap the pipe with a strip of paper and make the markings of pi*0D/4. You can make center hole, 90 hole, opposite hole with one shot. Done!

nonsense, punch anywhere and thats the center of pipe. when finished putting the intersecting pipe install with that pointing upward at 90 °.

Put the level in place of the square and measure off that
if you are going for the top or side of the pipe... just use a plumb bob... where it touches... center of pipe.

In your video tutorial at 02:07 you mentioned that 4 inch = 114 mm, however 1 inch = 25.4 mm therefore 4"= 101.6 mm.
